Germinación de embriones somáticos de Swietenia macrophylla en medios de cultivo semisólidos

With the objective to establish a suitable culture medium and to determine the optimum culture time for the germination of somatic embryos of Swietenia macrophylla King, it was carried out these work. Somatic embryos at the cotyledonal stage were placed in a culture medium formed by the half of the MS salts for their germination . three concentrations of 6-BAP (0.2, 0.4 and 0.6 mg.l-1), two concentrations of AG (1.0 and 2.0 mg.l-1) and a culture medium control without growth regulators were studied. In the culture medium wher3e the best results were obtained, the effect of the culture time on the germination of somatic embryos (30, 60 and 90 days of culture) was studied. The major number of somatic embryos with total germination was obtained in the culture medium without growth regulators, with significant differences to the rest of the treatments. The highest value of somatic embryos with secondary embryogenesis was presented when 0.6 mg.l-1 of 6-BAP were added to the culture medium. The increase of the culture time to 60 and 90 days increased significantly the total germination of the somatic embryos. These results allowed to define a culture medium for the germination of somatic embryos of Swietenia macrophylla King composed by the half of the MS salts without growth regulators with a culture time of 60 days.Key words: in vitro culture, Mahogany, regeneration, somatic embryogenesisCon el objetivo de establecer un medio de cultivo y determinar el tiempo de cultivo para la germinación de embriones somáticos de Swietenia macrophylla King, se realizó este trabajo. Embriones somáticos en etapa cotiledonal fueron colocados en un medio de cultivo compuesto por la mitad de las sales MS. Se estudiaron tres concentraciones de 6-BAP (0.2, 0.4 y 0.6 mg.l-1), dos concentraciones de AG3 (1.0 y 2.0 mg.l-1) y un medio de cultivo control sin reguladores de crecimiento. En el medio de cultivo que se obtuvieron los mejores resultados, se estudió el efecto del tiempo de cultivo (30, 60 y 90 días de cultivo) sobre la germinación de embriones somáticos. El mayor número de embriones somáticos con germinación total se obtuvo en el medio de cultivo sin reguladores de crecimiento con diferencias significativas con el resto de los tratamientos. Cuando se le adicionaron al medio de cultivo 0.6 mg.l-1 de 6-BAP se presentó el valor más alto de embriones somáticos con embriogénesis secundaria. El aumento del tiempo de cultivo a 60 y 90 días incrementó significativamente la germinación total de los embriones somáticos. Estos resultados permitieron definir un medio de cultivo para la germinación de embriones somáticos de Swietenia macrophylla compuesto por la mitad de las sales MS sin reguladores de crecimiento con un tiempo de cultivo de 60 días.Palabras clave: caoba, cultivo in vitro, embriogénesis somática, regeneració
