Bazı Gram Negatif Bakterilerin Bakteriyosin Üretimlerinin Araştırılması

In this study, strains of Escherichia coli, Pseudomonas aeruginosa and Klebsiella pneumoniae, which were isolated from several clinical samples, were investigated for their antibacterial activities. Strains of E. coli 26S, E. coli 14, E. coli 27, P. aeruginosa 8, P. aeruginosa 15, and P. aeruginosa 22 were found to have the highest activity. It was established that the antibacterial substances obtained from these strains were partially or completely sensitive to proteolytic enzymes. E. coli 26S and P. aeruginosa 8 were selected as the most efficient strains producing bacteriocin by disk diffusion method which was found to be the optimal method when spot-on-lawn, disc diffusion and well-diffusion methods were compared for determining antibacterial activity. Optimal bacteriocin production was established in Luria Bertani Broth medium at pH 7.0 and 37 oC in 21 hours for both strains. It was observed that agitated and static conditions did not affect the bacteriocin production.Bu çalışmada, çeşitli klinik örneklerden izole edilen Escherichia coli, Pseudomonas aeruginosa ve Klebsiella pneumoniae suşlarının antibakteriyel aktiviteleri araştırıldı. En yüksek antibakteriyel aktiviteye sahip suşlar E. coli 26S, E. coli 14, E. coli 27, P. aeruginosa 8, P. aeruginosa 15 ve P. aeruginosa 22 olarak bulundu. Bu suşlardan elde edilen antibakteriyel maddelerin proteolitik enzimlere kısmen ya da tamamen hassas olduğu saptandı. Antibakteriyel aktivite belirlenmesi için damlatma, disk difüzyon ve kuyucuk yöntemleri karşılaştırılarak en uygun yöntem seçilen disk difüzyon yöntemi ile bakteriyosin üreten en verimli suşlar olarak E. coli 26S ve P. aeruginosa 8 seçildi. Optimum bakteriyosin üretimleri her iki bakteri suşu için de Luria Bertani Broth besiyerinde, pH 7.0 de 37 oC de ve 21. saatte olduğu saptandı. Çalkalamalı ya da statik koşullar bakteriyosin üretimlerini etkilemedi
