Modulation of microtubule associated protein tau isoforms: rescue of functional and molecular phenotypes in a mouse model of tauopathy

Las enfermedades neurodegenerativas pueden ser causadas por una miríada de factores que conllevan a la muerte y/o disfunción de determinadas neuronas del cerebro humano. La enfermedad de Alzheimer (EA) y otras demencias similares se clasifican dentro del grupo de las taupatías, caracterizadas por el metabolismo anormal de la proteína tau asociada a los microtúbulos, la cual forma acumulaciones de depósitos insolubles neurofibrilares en las neuronas afectadas. La proteína Tau se expresa principalmente en neuronas, y en condiciones normales participa en la estabilización de los microtúbulos y en el transporte axonal. Los procesos patológicos que originan la formación de agregados de Tau no están completamente elucidados. En muchos casos se asocian a alteraciones postraduccionales de la proteína (por ejemplo, el estado de fosforilación) o a cambios en los procesamientos postranscripcionales del gen codificante para Tau (MAPT) (Lee et al., 2001). En particular, se han asociado taupatías con defectos en el procesamiento por splicing del ARN transcripto primario de Tau (Andreadis, 2012). En el cerebro humano adulto normal se detectan seis isoformas normales de Tau, derivadas del splicing alternativo de los exones 2, 3 y 10. El exón 10 (E10) en particular contiene un dominio clave de unión a los microtúbulos y su exclusión o inclusión genera dos isoformas de Tau presentes en cantidades equivalentes (conocidas como isoformas 3R y 4R) (Andreadis, 2005). En el gen MAPT, han sido identificadas más de 35 mutaciones asociadas con taupatías (Gallo et al., 2007); varias de las cuales no modifican la secuencia de la proteína, pero producen defectos en el mecanismo de splicing alternativo del E10, alterando el contenido relativo de las isoformas 3R y 4R de Tau en el cerebro de los pacientes afectados. En resumen, varias taupatías están asociadas con desequilibrios en las isoformas tau, por lo tanto, la restauración del equilibrio 3R:4R podría representar una terapéutica plausible. Aquí se utilizó una estrategia molecular de reprogramación del ARN por trans-splicing para modular la inclusión del E10 en la corteza prefrontal de un modelo de ratón (hTau) de taupatía, el cual produce un exceso anormal de la isoforma 3R de tau. Luego de la ocurrencia del evento de trans-splicing, en los ratones tratados se alcanzó un equilibrio parcial 3R:4R, con una reducción significativa de tau insoluble e hiperfosforilada. Además, la activación neuronal y el comportamiento de estos ratones se restauraron a los niveles del grupo WT. Por otra parte, utilizando la misma estrategia de trans-splicing logramos modular la inclusión/exclusión del E10 de tau en neuronas humanas diferenciadas en cultivo, derivadas de células madre. Con los cambios en el contenido relativo de tau 3R: 4R, las neuronas no mostraron cambios morfológicos,sin embargo, los análisis de trayectorias individuales de las vesículas móviles revelaron trayectorias en el transporte axonal alteradas. En conjunto, nuestros resultados revelan que la modulación temprana del splicing del E10 en la transcripción de tau puede rescatar defectos fenotípicos relevantes asociados con el splicing incorrecto de tau, lo que plantea perspectivas prometedoras para el uso de la reprogramación de ARN para taupatías humanas.Neurodegenerative diseases can be caused by a myriad of factors that lead to death or dysfunction of certain neurons in the human brain. Alzheimer's disease (AD) and other similar dementias are classified within the group of tauopathies, which are characterized by the abnormal metabolism of the tau microtubule-associated protein, leading to accumulations of insoluble neurofibrillary deposits in the affected neurons. Tau protein is mainly expressed in neurons, and under normal conditions participates in the stabilization of microtubules and axonal transport. The pathological mechanism that cause the formation of Tau aggregates are not completely elucidated. In many cases, they are associated with posttranslational alterations of the tau protein (for example, the state of phosphorylation) or changes in the posttranscriptional processes of the gene coding for Tau (MAPT) (Lee et al., 2001). In particular, the tauopathies have been associated with defects in the processing by splicing of the primary transcribed RNA of MAPT (Andreadis, 2012). In the adult human brain, six normal Tau isoforms are detected, derived from the alternative splicing of exons 2, 3 and 10. The exon 10 (E10) in particular encodes the second of four microtubule-binding repeats and its exclusion or inclusion by splicing gives rise to tau isoforms with 3 or 4 repeats, present in equal amounts in the normal adult human brain (known as 3R and 4R isoforms) (Andreadis, 2005). In the MAPT gene, more than 35 mutations associated with tauopathies have been identified (Gallo et al., 2007); most of them do not modify the protein sequence, but produce defects in the alternative splicing mechanism of E10, altering the relative content of the tau 3R and 4R isoforms in the brain of affected patients. In summary, several taupathies are associated with imbalances in tau isoforms, therefore, the restoration of the 3R: 4R equilibrium could represent a plausible therapeutic approach. Here we used a molecular strategy of RNA reprogramming by trans-splicing to modulate the inclusion of E10 in the prefrontal cortex (PFC) of a mouse model (hTau) of tauopathy, which produces an abnormal quantity of the 3R tau isoform. After the trans-splicing event, a ratio 3R:4R partial equilibrium was achieved in the PFC of treated mice, with a significant decreased of insoluble and hyperphosphorylated tau. In addition, neuronal firing and behavior in this group were restored to the levels of the WT mice. On the other hand, using the same trans-splicing strategy we modulated the inclusion/exclusion of tau E10 in human differentiated neurons derived from stem cells. With changes in the relative content of tau 3R: 4R, neurons did no develop morphological changes; however, single trajectory analyses of the moving vesicles showed impairments in axonal transport. Taken together, our results reveal that early modulation of E10 splicing in tau transcription can rescue relevant phenotypic defects associated with incorrect splicing of tau, which raises promising prospects for the use of RNA reprogramming for human tauopathies.Fil: Espindola, Sonia Lorena.
