Effectiveness of lemon tea tree essential oil against enterococcus faecalis biofilm on bovine dentin

Aim: The aim of this study was to evaluate and compare, ex vivo, the effectiveness of calcium hydroxide [Ca(OH)2] and Lemon Tea Tree essential oil (LTT) against E. faecalis biofilm on root canal of bovine teeth. Methods: After E. faecalis biofilm formation in 42 dentin tubes of bovine teeth, the specimens were randomly divided according to the intracanal dressing (n=10): G1- Ca(OH)2; G2- LTT 5%; e G3- control - saline 0.85%. After 7 days, intracanal medication was removed and the collection of microbiological samples from the remaining biofilm was performed. The dentin tubes without medication were immediately immersed in sterile culture medium and stored for 7 days to allow the analysis of residual antimicrobial effect of each medication. After that, another microbiological sampling was performed. The bacterial viability test, by colony forming units (CFUs) counting, was carried out from the material collected in both moments. The architecture of biofilms of 4 specimens of each group were analyzed by scanning electron microscopy. The mean values of CFUs were analyzed by one way ANOVA and post hoc Tukey tests (P < 0.05). Results: Results obtained after removal of medications showed a 100% microbial reduction promoted by Ca(OH)2 (P < 0,001) and 96,31% promoted by LTT 5% (p < 0,004), compared to the control. Only Ca(OH)2 showed significant residual antimicrobial effect (P = 0,001), although LTT 5% had eliminated 69,89% of E. faecalis cells. Conclusion: The LTT 5%, as intracanal dressing, showed excellent antimicrobial effectiveness, comparable to the Ca(OH)2, against E. faecalis biofilm on root canal of bovine teeth.Objetivo: O objetivo deste estudo foi avaliar e comparar, ex vivo, a efetividade do hidróxido de cálcio [Ca(OH)2] e do óleo essencial Lemon Tea Tree (LTT) contra o biofilme de E. faecalis estabelecido no canal radicular de dentes bovinos. Material e Métodos: Após a formação do biofilme de E. faecalis em 42 tubos de dentina de dentes bovinos, os espécimes foram divididos aleatoriamente de acordo a medicação intracanal empregada (n=10): G1- Ca(OH)2; G2- LTT 5%; e G3- controle - solução salina 0,85%. Passados 7 dias, as medicações foram removidas e a coleta microbiológica do biofilme remanescente foi realizada. Os tubos de dentina sem as medicações foram imediatamente imersos em meio de cultura estéril e armazenados por mais 7 dias para a análise do efeito antimicrobiano residual das medicações. Após este período, foi feita uma nova coleta microbiológica. Do material coletado nos dois momentos, foi realizado o teste de viabilidade bacteriana, por meio da contagem de Unidades Formadoras de Colônias (UFCs). Quatro espécimes de cada grupo foram submetidos à análise em microscopia eletrônica de varredura para observação da arquitetura do biofilme. Os valores médios de CFUs foram analisados pelos testes one way ANOVA e post hoc Tukey (P < 0,05). Resultados: Logo após a remoção da medicação, o Ca(OH)2 demonstrou uma redução de 100% das células microbianas (P < 0,001) e o óleo LTT 5%, de 96,31% (p < 0,004), comparados ao controle. Apenas o Ca(OH)2 apresentou efeito antimicrobiano residual significativo (P = 0,001), ainda que o LTT 5% tenha eliminado 69,89% das células de E. faecalis. Conclusão: O LTT 5%, empregado na forma de medicação intracanal, apresentou excelente efetividade antimicrobiana, comparável ao Ca(OH)2, contra o biofilme de E. faecalis estabelecido na dentina do canal radicular de dentes bovinos
