The collected water samples are then stored in a box filled with sufficient ice to keep the sample temperature below 4°C during transit. When we arrived in the laboratory, we used microporous membrane (0.22 m) to filter water samples, filtered pore size filter membrane, stored in -80°C refrigerator, DNA was extracted with Water DNA Kit kit from OMEGA company, The extracted DNA was amplified using a specific set of bar-code 515F and 909R.
