Assessment of Copper Sulphate Impact on Cloning Dynamics and Functional State of the Benthic Diatoms (Bacillariophyta)

Many species of benthic diatoms (Bacillariophyta) are low-resistant to ecological stressors and, therefore, changes in its development features under various toxicants’ impact can be considered as indirect assessment of the environment quality. The study is focused on investigation of the cloning dynamics of diatom cells in stock culture under single addition of copper sulfate (CuSO4) in five successively decreasing concentrations (up 500 mkg l-1 to 31.3 mkg l-1). The clone cultures of 4 marine benthic diatom species Psammodictyon panduriforme var. continua (Grunow) Snoeijs 1998, Entomoneis paludosa (W. Smith) Reimer in Patrick & Reimer 1975, Cyclophora tenuis Castracane 1878 and Haslea sp. were used in experiments. By the results of 10-days toxicological experiments was ascertain that according to resistance to the toxicant the considered species are split into two groups. The high sensitive species P. panduriforme var. continua и Haslea sp. stopped cell division process already at the minimum concentration of toxicant (31.3 mkg l-1). Toxic benchmark of the copper sulphate causing to termination of growth and development of cell number for these 2 species is very low and is within range 0 to 31 mkg l-1. For moderately tolerant species E. paludosa 62 mkg l-1 is the threshold concentration of CuSO4 when differences in average cell number comparing with the control level are statistically significant under various exposure time of experiments. For tolerant C. tenuis the threshold concentration of CuSO4 is 125 mkg l-1 by which depression of cell growth are always significantly differed from the control. By low concentrations of toxicant (31 and 62 mkg l-1), increasing the cell number of moderately tolerant species along to extension of experiments can be well extrapolated by power function (for C. tenuis) or by sigmoid function (for E. paludosa). In the control, vegetative reproduction of all 4 investigated diatom species can be approximated by the exponential curve fitting for different duration of experiments (3 to 10 days). The certain peculiarities of diatom cells physiological response under high concentration of CuSO4 were marked, such as abnormality of morphogenesis and multiple non-separations of valves from any one side of cell after vegetative phase of cytokinesis.Многие виды диатомовых слаборезистентны к влиянию экологических стрессоров, поэтому изменения показателей их развития под воздействием различных токсикантов могут быть применены как опосредованная оценка качества среды. Целью работы было исследование динамики вегетативного деления клеток диатомовых водорослей при однократном внесении раствора сульфата меди (CuSO4) в пяти последовательно снижающихся концентрациях (от 500 мкг·л-1 до 31.3 мкг·л-1). В эксперименте использованы клоновые культуры четырёх видов морских бентосных форм: Psammodictyon panduriforme var. continua (Grunow) Snoeijs 1998, Entomoneis paludosa (W. Smith) Reimer in Patrick & Reimer 1975, Cyclophora tenuis Castracane 1878 и Haslea sp. По результатам 10-суточных токсикологических экспериментов установлено, что по степени резистентности к токсиканту изученные виды подразделяются на две группы. Высокочувствительные виды P. panduriforme var. continua и Haslea sp. прекращают деление клеток уже при минимальных концентрациях токсиканта (31.3 мкг·л-1). Токсический порог воздействия сульфата меди, вызывающий прекращение развития и роста численности клеток, у данных двух видов очень низкий (диапазон между контролем и концентрацией 31.3 мкг·л-1). В группе среднетолерантных видов для E. paludosa пороговой является концентрация сульфата меди 62.5 мкг·л-1, при воздействии которой различия в средних значениях численности клеток всегда статистически достоверны. Для C. tenuis пороговая концентрация CuSO4 составляет 125 мкг·л-1, при которой угнетение роста клеток относительно контроля всегда статистически достоверно. При низких концентрациях токсиканта (31.2 и 62.5 мкг·л-1) рост численности клеток среднетолерантных видов по мере увеличения периода экспозиции аппроксимируется степенной (для C. tenuis), либо затухающей сигмоидной моделью (для E. paludosa). В контроле у всех четырех исследованных видов диатомовых вегетативное размножение клеток идет по экспоненциальной модели на протяжении эксперимента. Отмечены особенности физиологического ответа клеток диатомовых при высокой концентрации токсиканта, выраженные в нарушении процесса морфогенеза и множественном нерасхождении створок клеток с одной стороны после вегетативной фазы цитокинеза
