Breast and ovarian cancers are common cancers affecting women. Family history of the disease is a major risk factor for both cancers. Even though several susceptibility alleles have been identified, genetic predisposition to breast and ovarian cancer is still largely unexplained. Most of the known risk-genes play a role in DNA repair and especially in homologous recombination repair. RAD51C, RAD51D, and RAD54L are genes involved in homologous recombination and rare mutations in RAD51C and RAD51D have been identified in breast and ovarian cancer families.
