Evaluation of the potential on the removal of heavy metals and/or pesticides by actinobacteria and plant species grown in co-contaminated systems

La co-contaminación ambiental es un problema que enfrenta actualmente la población mundial debido al avance tecnológico y al crecimiento poblacional. Las técnicas de fitorremediación, basadas en las interacciones entre plantas y microorganismos se han propuesto como métodos rentables y ecológicos para limpiar sitios contaminados con tóxicos orgánicos e inorgánicos. Varios estudios demuestran la elevada disipación de contaminantes en la interfase suelo-raíz, atribuida principalmente al aumento en la densidad, diversidad y/o actividad metabólica microbiana debido a la liberación de exudados radiculares vegetales. Estas asociaciones plantas-microorganismos pueden mejorar significativamente los procesos de fitorremediación del suelo promoviendo la salud y el crecimiento vegetal, y modificando la movilidad y disponibilidad de los contaminantes. En este contexto, el objetivo de esta Tesis Doctoral fue evaluar la remoción de metales pesados y/o plaguicidas por actinobacterias y especies vegetales cultivadas en sistemas co-contaminados. Se aislaron actinobacterias desde muestras de suelo, rizósfera y especies vegetales desarrolladas en un sitio desde el que se había removido un depósito clandestino de plaguicidas. Se seleccionaron 57 colonias microbianas por presentar características compatibles con las de actinobacterias; se realizaron ensayos cualitativos de tolerancia microbiana a Cr(VI) y/o lindano, a partir de los que se seleccionaron cinco cepas en virtud de su crecimiento en presencia de ambos contaminantes. Se realizó la caracterización molecular de estos aislamientos denominados Z38, Z2, ZF, ZII y ZI, como así también el estudio de sus propiedades promotoras del crecimiento vegetal y se determinó mediante secuenciamiento del ADNr 16S su filiación con cepas del género Streptomyces. Ensayos cuantitativos en medio liquido permitieron determinar que Streptomyces sp. Z38 y Z2 presentaban los mayores valores de remoción de Cr(VI) y lindano en medio de cultivo co-contaminado, por lo que se evaluó posteriormente el desempeño de ambos microorganismos en medio co-contaminado suplementado con exudados radiculares (ERs) de plantas de maíz. Comparando dicha fuente de carbono con otra como la glucosa, se encontró que la disipación de Cr(VI) en medio co-contaminado mejoraba considerablemente en presencia de ERs. En un ensayo posterior se incrementó la concentración de lindano en el medio co-contaminado y se observó que la remoción del plaguicida continuaba siendo elevada en dicho sistema, en contraposición a lo que sucedía con el cromo cuya remoción disminuyó significativamente. Debido a que Streptomyces sp. Z38 presentó propiedades promotoras del crecimiento vegetal, así como también características morfológicas y fisiológicas propicias para crecer en suelo, fue seleccionada para evaluar la remoción de Cr(VI) y/o lindano en suelo artificialmente contaminado, implantado con maíz e inoculado con la bacteria. La mayor remoción de lindano se obtuvo en el sistema implantado e inoculado, mientras que la mayor remoción de Cr(VI) se obtuvo en el sistema implantado (sin microorganismo) y en el sistema inoculado (sin planta). Los resultados de estudios fisiológicos realizados en las plantas sugieren que Streptomyces sp. Z38 induciría un efecto protector en las mismas frente al metal. Mediante un ensayo de ecotoxicidad se demostró la disminución de Cr(VI) y lindano en los suelos tratados.Environmental co-contamination is a problem that currently faces the world wide population due mainly to technological progress and population growth. Phytoremediation techniques, based on the interactions between plants and microorganisms, have been proposed as cost-effective and ecological methods to clean up sites contaminated with organic and inorganic contaminants. Several studies have shown higher dissipation of contaminants at the root-soil interface, attributed mainly to the increase in density, diversity and/or microbial metabolic activity due to the release of plant root exudates. These plants-microorganisms interactions can significantly improve soil phytoremediation processes, promoting plant health and growth, and modifying the mobility and availability of contaminants. In this context, the objective of this Doctoral Thesis was to evaluate the removal of heavy metals and/or pesticides by actinobacteria and plant species grown in co-contaminated systems. Actinobacteria were isolated from soil samples, rhizosphere and plant species developed in a site from which a clandestine deposit of pesticides had been removed. Fifty-seven microbial colonies were selected because they had characteristics compatible with those of actinobacteria. Qualitative tests of microbial tolerance to Cr(VI) and/or lindane were carried out, from which five strains were selected according to their growth in the presence of both contaminants. The molecular characterization of these isolates called Z38, Z2, ZF, ZII and ZI was carried out, as well as the study of their plant growth promoting properties. By sequencing of the 16S rDNA gene, strains were identified as members of the Streptomyces genus. Quantitative assays in a liquid medium allowed to determine that Streptomyces sp. Z38 and Streptomyces sp. Z2 had the highest values of Cr(VI) and lindane removal in co-contaminated culture medium, so the performance of both microorganisms in a co-contaminated medium supplemented with maize root exudates (REs) was evaluated. Comparing REs with another carbon source such as glucose, it was found that the dissipation of Cr(VI) in co-contaminated medium improved considerably in the presence of REs. In another assay, the concentration of lindane in the co-contaminated medium was increased and it was observed that lindane dissipation remained high in this system, in opposite to what happened with the chromium whose removal decreased significantly. Because Streptomyces sp. Z38 presented several plant growth promoting properties, as well as morphological and physiological characteristics favourable to grow in soil, it was selected to evaluate the removal of Cr(VI) and/or lindane in artificially co-contaminated soil implanted with maize and inoculated with the bacteria. The largest lindane removal was obtained in the implanted and inoculated system, while the highest Cr(V I) removal was obtained in the implanted system (without microorganism) and in the inoculated system (without plant). The results of physiological studies carried out on the plants suggest that Streptomyces sp. Z38 would induce a protective effect on the plant.
