There are many open questions on the motion, functioning and control of eukaryotic cilia, including their hydrodynamic interactions with water, and the dynamics of their intracellular signaling. Studies have been limited by the lack of reliable data on their three dimensional motion and the inability to record data for sustained periods. This dissertation addresses the problem with new experimental techniques and instruments to facilitate studies on ciliary motility. The objective of this work is to build a reliable real-time imaging system capable of recording and processing cellular behavior on the time scale of bio-chemical reactions that take place within the cell.
