The thesis investigated mitochondrial function during the development of insulin resistance in skeletal muscle and fatty liver by feeding a high fat diet in different age groups of C57BL/6J mice. In skeletal muscle mitochondria performed normally, but in liver mitochondrial ROS production induced a Nrf2-mediated response. A reduction of lysosomal proteins in middle-aged mice impaired mobilization of triglycerides contributing to fat accumulation together with reduced fatty acid oxidation
