This thesis deals with the analysis of tobacco as well as mainstream and sidestream smoke of cigarettes by means of time-of-flight mass spectrometry (TOFMS) with soft ionisation methods (single photon ionisation, (SPI), resonance enhanced multi photon ionisation (REMPI)) as well as comprehensive two-dimensional gas chromatography (GCxGC). With a newly developed smoking machine, which is capable of online-analysis, the influence of several smoking parameters on the chemical composition of mainstream smoke was investigated. Utilising statistical methods it was shown that the amount of several toxic compounds and the chemical composition of the smoke is strongly dependant on puff duration and puff interval.
