The restless legs syndrome (RLS) is a common complex genetic neurological disorder. In this work, the first large-scale SNP-based case-control association studies of RLS identified associations to intronic and intergenic SNPs in five genomic regions. Regional studies of known linkage regions found associated variants in the genes NOS1 and PTPRD. A genome-wide study identified associated variants in three genomic regions containing the genes MEIS1, BTBD9, MAP2K5, and LBXCOR1. Each variant confers a 30 to 70% increase in disease risk and a haplotype in MEIS1 leads to an almost 3-fold increase.
