Orf Virus (ORFV) is a large, 134 kbp dsDNA virus that is the type species of the Parapoxvirus genus. ORFV infections are epitheliotropic resulting in lesions of the skin. The natural host range of the virus consists of sheep and goats, although zoonotic infection of humans is common. ORFV encodes several immuno-modulating factors that allow this virus to manipulate a range of critical host antiviral defence mechanisms. One of the most important mammalian signalling pathways is the JAK/STAT pathway, stimulated by IFN-α/β and IFN-γ. Stimulation of this pathway results in the upregulation of hundreds of genes involved in the innate and adaptive antiviral immune response.
