Revitalisation of mammalian cells exposed to antiseptics

The cytotoxic activity of the antiseptics octenidine dihydrochloride (OCT), polihexanid (PHMB), and povidone iodine (PVP-I) were investigated on epithelial cells (CHO-K1, ATCC CCL61) and fibroblasts (L929, ATCC CCL1) both immediately after 30 min exposure and after 30 min exposure followed by a culture after 24 h without active agents and compared with chlorhexidine digluconate (CHX), and silver-containing formulations. The cytotoxic reaction was detected using MTT assay (intracellular cytotoxicity) and neutral red (NR) assay (damage of the cell membrane). Concentrations of PVP-I resulting after 30 min in >20% vitality revitalize after 24 h of culture without PVP-I. The cytotoxic effect of PHMB is only partially reversible in the cell membrane cytotoxicity at concentrations below the IC50 (vitality >50%). A total revitalization was impossible, because simutaneously the intracellular cytotoxicity increases during the 24 h culture. The cytotoxic effect of OCT, CHX, and silver-containing formulations is irreversible, but only the cell-bound OCT remains biologically activity against microorganisms.Die antiseptischen Wirkstoffe Octenidindihydrochlorid (OCT), Polihexanid (PHMB) und Polyvinylpyrrolidon-Iod (PVP-I) wurden gegenüber Epithelzellen (CHO-K1, ATCC CCL61) und Fibroblasten (L929, ATCC CCL1) hinsichtlich ihrer unmittelbaren (nach 30 min Kontakt) und nachfolgenden (auf die 30 min Kontakt folgende 24 h Folgekultur ohne Wirkstoff) zytotoxischen Reaktion im Vergleich zu Chlorhexidindigluconat (CHX) und silberhaltigen Präparaten untersucht. Als Nachweisverfahren wurden der MTT-Test (intrazelluläre Zytotoxizität) und der Neutralrot (NR)-Test (Zellmembranschädigung) verwendet. Konzentrationen von PVP-I, die bei den Prüfzellen nach 30 min Exposition einen Vitalitätserhalt >20% gewährleisten, revitalisieren während der 24 h Folgekultur ohne Wirkstoff vollständig. Die zytotoxische Wirkung von PHMB ist nur für die Zellmembran-Zytotoxizität bei Konzentrationen unterhalb der IC50 teilweise reversibel. Eine vollständige Revitalisierung ist jedoch nicht möglich, da gleichzeitig die intrazelluläre zytotoxische Wirkung von PHMB zunimmt. Die zytotoxische Wirkung von OCT, CHX und silberhaltigen Präparaten ist irreversibel. Der von den Zellen gebundene Wirkstoff bleibt gebunden und verstärkt während der 24 h Folgekultur seine zytotoxische Wirkung. Zellgebundenes OCT besitzt jedoch gegenüber zellgebundenem CHX den Vorteil, dass es gegenüber Mikrorganismen biologisch aktiv bleibt
