In the context of transdermal drug delivery systems one aspect still is difficult to access. This is the determination of the saturation solubility of a drug in a polymeric matrix and associated with this the influence of the thermodynamic activity on the flux from the TDSs. In this thesis both problems are adressed. First, the 5-layer laminate technique for the determination of the saturation solubility of a drug in a polymeric matrix was substantially refined to enable a fast determination of the solubility. With this improved method, the saturation solubility of scopolamine in three acrylic adhesives was determined at room temperature and 45 °C, and in the presence of different additives.
