Systematic Analyses of Combination Effects of HHV-8-encoded Genes on NF-kappaB Activation

Das Humane Herpesvirus 8 (HHV-8) ist das ursächliche Agens des Kaposi-Sarkoms (KS) und des Primären Effusionslymphoms (PEL). Bei der Pathogenese des KS und bei der Entstehung des PEL wird dem zellulären Transkriptionsfaktor nukleärer Faktor-kappa B (NF-kappaB) eine wesentliche Rolle zugeschrieben. Die Aktivierung von NF-kappaB ist notwendig für die latente Persistenz von HHV-8, für das Überleben der HHV 8-infizierten Zellen und für das Fortschreiten HHV-8-assoziierter Krankheiten. In dieser Arbeit wurde die Methode der reversen Transfektion (Synonym: Zellchip-Analyse) benutzt, um systematisch den Einfluss von HHV-8-Genen auf den NF-kappaB-Signalweg zu untersuchen. Hierzu wurden alle HHV-8-Gene einzeln (n = 86) und zusätzlich alle paarweisen Kombinationen der K-Gene und latenten Gene (n = 231) eingesetzt, um Aktivatoren oder Hemmstoffe des NF-kappaB-Signalweges zu identifizieren. Die statistische Auswertung von mehr als 14.000 Transfektionen erbrachte ORF75 als einen neuen und bestätigte K13 als einen bekannten HHV-8-kodierten Aktivator von NF-kappaB. K13 und ORF75 kooperierten in der Aktivierung von NF-kappaB. Eine Hemmung der ORF75-Expression durch spezifische siRNA zeigte, dass ORF75 signifikant an der NF-kappaB-Aktivierung in HHV-8-infizierten Zellen beteiligt ist. Zusätzlich wurde K10.5 als NF-kappaB-Inhibitor bestätigt und K1 als neuer Hemmstoff der K13- und ORF75-induzierten NF-kappaB-Aktivierung identifiziert. Alle durch Zellchip-Analysen ermittelten Ergebnisse konnten durch klassische Transfektionsexperimente bestätigt werden. Diese Arbeit beschreibt die erste erfolgreiche Anwendung der Zellchip-Analyse zur systematischen Untersuchung der Pathofunktionen von Genen eines infektiösen Agens. Mit dieser Methode wurden ORF75 und K1 als neue HHV-8-kodierte Modulatoren des NF-kappaB-Signaltransduktionsweges identifiziert. Die ermittelten Gene sind möglicherweise in der Feinabstimmung des Gleichgewichts zwischen latenten und lytischen Lebenszyklus beteiligt, da dieses Gleichgewicht entscheidend vom Zustand der NF-kappaB-Aktivierung abhängig ist.Human herpesvirus 8 (HHV-8) is the etiologic agent of Kaposi’s sarcoma and primary effusion lymphoma. Activation of the cellular transcription factor nuclear factor-kappa B (NF-kappaB) is essential for latent persistence of HHV-8, survival of HHV-8-infected cells and disease progression. We used reversely transfected cell microarrays (RTCM, synonymously cell chip analysis) as an unbiased systems biology approach to analyze systematically the effects of HHV-8 genes on the NF-kappaB signaling pathway. To this goal all HHV-8-encoded genes individually (n = 86) and additionally all K- and latent genes in pairwise combinations (n = 231) were investigated in order to detect activators or inhibitors of NF-kappaB. Statistical analyses of more than 14,000 transfections identified ORF75 as a novel and confirmed K13 as a known HHV-8-encoded activator of NF-kappaB. K13 and ORF75 showed cooperative NF-kappaB activation. siRNA-mediated knockdown of ORF75 expression demonstrated that this gene significantly contributes to NF-kappaB activation in HHV-8-infected cells. Furthermore, our approach confirmed K10.5 as an NF-kappaB inhibitor and newly identified K1 as an inhibitor of both K13- and ORF75-mediated NF-kappaB activation. All results obtained with RTCM were confirmed with classical transfection experiments. Our work describes the first successful application of RTCM for the systematic analysis of pathofunctions of genes encoded by an infectious agent. With this approach ORF75 and K1 were identified as novel HHV-8-encoded regulatory molecules on the NF-kappaB signal transduction pathway. The identified genes may be involved in fine-tuning of the balance between latency and lytic replication since this depends critically on the state of NF-kappaB activity
