Analyse der Rolle der zellulären subnukleären Struktur ND10 für die humane Cytomegalovirus Replikation

Human cytomegalovirus (HCMV) infection commonly results in the association of viral genomes and important regulatory proteins like pp71, IE1, or IE2 with a cellular subnuclear structure known as the nuclear domain 10 (ND10). Since only ND10-associated genomes initiate viral gene expression, ND10 domains were hypothesized to be essential for viral infection. On the other hand, many ND10 factors like PML, hDaxx, or Sp100 are interferon-inducible and ND10 accumulations are disrupted by the IE1 protein during the course of viral infection, suggesting that ND10 could also be part of an intrinsic defense mechanism of the cell. In order to determine the relevance of ND10 domains for HCMV replication, primary human fibroblasts (HFFs) with a stable, siRNA-mediated knockdown (kd) of the major ND10 components PML and hDaxx were generated. Comparative analysis of HCMV infection in PML- and hDaxx-kd cells revealed a similarly increased initiation of viral gene expression, regardless of which ND10 constituent was depleted. Since loss of PML, the defining component of ND10, results in a dispersal of the entire nuclear substructure, the enhanced replication efficacy of HCMV in PML-kd cells could also be a consequence of the dissociation of the repressor protein hDaxx from its optimal subnuclear site. However, several experiments were performed to demonstrate that PML and hDaxx independently contribute to the silencing of HCMV gene expression. Thus, this study identified the ND10 constituents PML and hDaxx as cellular restriction factors mediating intrinsic immunity against HCMV infection. This further supports the evolving concept of ND10 as part of an antiviral defense mechanism of the cell. Moreover, experiments with HCMV deletion mutants indicated that the viral regulatory factors pp71 and IE1 specifically antagonize hDaxx- or PML-mediated repression. Given the antiviral function of ND10, the in vivo association of the viral transactivators pp71 and IE2 with ND10 was analyzed in detail using recombinant HCMVs expressing fluorescently labeled versions of both regulatory proteins. Interestingly, insertion of an EYFP-pp71 coding sequence into the HCMV genome yielded a recombinant virus with enhanced growth properties compared to the wild-type virus due to increased amounts of pp71 present after infection. Initial experiments suggest that as yet unidentified functions of pp71 could account for the augmented replication efficiency of the EYFP-pp71 virus. Live cell imaging studies using mCherry-Sp100/-PML expressing cells for infection with the recombinant viruses indicated that incoming viral components are rapidly targeted by ND10 followed by a very dynamic spatial association between viral nucleoprotein complexes and ND10. While the hDaxx protein was identified as the decisive determinant for the subnuclear localization of pp71 at ND10, viral DNA turned out to be responsible for the dot-like IE2 accumulations, which form adjacent to ND10 during infection. Studies conducted with a DNA-binding-deficient IE2 mutant implied, that the formation of viral nucleoprotein complexes may constitute a critical step during the immediate early phase of a lytic infection.Infektionen mit humanem Cytomegalovirus (HCMV) führen generell zu einer Assoziation viraler Genome und Regulatorproteine wie pp71, IE1 oder IE2 mit einer als nukleäre Domäne 10 (ND10) bezeichneten subnukleären Struktur. Da nur ND10-lokalisierte Genome die virale Genexpression initiieren, wurde eine essentielle Funktion dieser Domäne für die virale Replikation vermutet. Andererseits ist bekannt, dass einige ND10-Faktoren wie PML, Sp100 und hDaxx Interferon-induzierbar sind und ND10-Akkumulationen im Verlauf der Infektion, vermittelt durch das IE1-Protein, aufgelöst werden, so dass bei ND10 auch über eine antivirale Funktion spekuliert wurde. Um die Relevanz von ND10 für die HCMV-Replikation aufzuklären, wurden primäre humane Fibroblasten mit einem stabilen, siRNA-vermittelten knockdown (kd) der ND10-Proteine PML und hDaxx generiert. Infektion der PML- und hDaxx-kd Zellen ergab eine in gleicher Weise gesteigerte Initiation der HCMV-Genexpression. Da PML die strukturgebende Komponente von ND10-Domänen darstellt, könnte die verstärkte Replikationseffizienz von HCMV in PML-kd Zellen auch auf eine ND10-Dissoziation des Repressorproteins hDaxx zurückzuführen sein. Aufgrund weiterführender Analysen konnte jedoch eindeutig eine unabhängige Beteiligung von PML und hDaxx an der Repression der HCMV Genexpression nachgewiesen werden. Somit wurden in dieser Arbeit PML und hDaxx als zelluläre Restriktionsfaktoren identifiziert, die eine intrinsische Immunität gegenüber HCMV Infektionen vermitteln. Zudem konnte gezeigt werden, dass die viralen Regulatorproteine pp71 und IE1 die hDaxx- bzw. PML-vermittelte Repression spezifisch antagonisieren. Des Weiteren wurde die ND10-Assoziation der viralen Transaktivatoren pp71 und IE2 in vivo mittels rekombinanter Viren, die Fluoreszenz-markierte Varianten beider Proteine exprimieren, eingehender untersucht. Interessanterweise resultierte die Insertion einer EYFP-pp71 Fusionssequenz in das HCMV-Genom in einer gesteigerten Infektiosität des rekombinanten im Vergleich zum Wildtyp-Virus; dies korrelierte mit einer erhöhten pp71-Proteinmenge im Kontext der Infektion. Initiale Experimente deuten darauf hin, dass eine noch unbekannte Funktion von pp71 zu der gesteigerten Replikationseffizienz des EYFP-pp71 Virus beiträgt. Lebendzell-Mikroskopiestudien unter Verwendung von mCherry-Sp100/-PML exprimierenden Zellen ergaben, dass virale Komponenten nach Zellkerneintritt rasch von ND10-Faktoren lokalisiert werden, gefolgt von einer sehr dynamischen Assoziation zwischen viralen Nukleoproteinkomplexen und ND10. Während das hDaxx-Protein als entscheidender Faktor für die Lokalisation von pp71 an ND10 identifiziert wurde, ist die Ausbildung ND10-assoziierter IE2-Akkumulationen während der Infektion auf eine IE2-Interaktion mit viraler DNA zurückzuführen. Experimente mit einer DNA-bindungsdefizienten IE2-Mutante deuten darauf hin, dass die Ausbildung derartiger Nukleoproteinkomplexe entscheidend ist für die initiale Phase der lytischen Infektion
