The enzyme ATP(CTP):tRNA nucleotidyltransferase is essential in eukaryotes to allow tRNAs to participate in protein synthesis. It adds a specific cytidine-cytidine-adenosine (CCA) sequence to generate positions 74, 75, and 76 at the 3' ends of tRNAs in a nucleic acid template-independent manner. Considerable effort has gone into exploring the catalytic mechanism of this enzyme and several bacterial forms, as well as the human enzyme, have been crystallized either in the presence or absence of substrates.
