Investigation of placental markers correlated with preeclampsia in second and third trimester pregnancies

Introduction: Preeclampsia is a serious obstetric problem and is characterized by significant rates of maternal and neonatal morbidity and mortality and its cause remains virtually unknown despite the several theories that have been put forward.Objective of the study: The investigation of 6 placental tissue markers and the comparison of their expression in placentas between preeclamptic women and women who did not develop preeclampsia. Because of the correlation of abnormal placentation which occurs in preeclampsia with cell apoptosis, we studied the markers TUNEL, C8 and M30, and because of the association of abnormal placentation with the process of tumorgenesis we studied the markers MTA1, MTA3 and CRIP1.Materials: The materials for the execution of this experiment were fifty-one (51) samples of placental tissue. Τwenty four (24) of them were derived from tissues of preeclamptic pregnancies while the rest twenty-seven (27) were obtained from normal pregnancies and were used as controls.Experimental procedure: The samples after the procedure of immunohistochemistry were studied by electron microscopy, and their expression was scored according to the German semi-quantitative scoring system considering the staining intensity and extent (PPSI).Conclusions: The expression values (PPSI) of all 6 markers was significantly higher in preeclamptic patients compared to the control group (p <0,001). The statistically significant difference in the expression of these placental indicators between pregnancies of preeclampsia and those without preeclampsia suggests that they play an important role in the evolution of preeclampsia and that will be a useful tool for further research in the prevention and prognosis of such a serious pathological entity for the pregnant woman and the fetus.Εισαγωγή: Η προεκλαμψία είναι ένα σοβαρό μαιευτικό πρόβλημα και χαρακτηρίζεται από σημαντικά ποσοστά μητρικής και νεογνικής νοσηρότητας και θνητότητας και η αιτία της παραμένει ουσιαστικά άγνωστη παρά τις διάφορες θεωρίες που έχουν διατυπωθεί.Στόχος μελέτης: Η διερέυνηση 6 δεικτών πλακουντιακού ιστού και η σύγκριση της έκφρασης τους σε πλακούντες μεταξύ προεκλαμπτικών γυναικών και γυναικών που δεν εμφάνισαν προεκλαμψία. Λαμβάνοντας υπόψη την συσχέτιση της παθολογικής πλακουντοποίησης που επισυμβαίνει στην προεκλαμψία με την κυτταρική απόπτωση μελετήθηκαν οι δείκτες ΤUNEL, C8 και Μ30 και λόγω της συσχέτισης της παθολογικής πλακουντοποιησης με την διαδικασία της ογκογένεσης μελετήθηκαν οι δείκτες ΜΤΑ1, ΜΤΑ3 και CRIP1.Υλικά: Το υλικό για την εκτέλεση του πειράματος αυτού αποτέλεσαν πενήντα ένα (51) δείγματα πλακουντιακών ιστών. Τα είκοσι τέσσερα (24) από αυτά προήλθαν από ιστούς προεκλαμπτικών κυήσεων ενώ τα υπόλοιπα είκοσι εφτά (27) προήλθαν από φυσιολογικές κυήσεις και χρησιμοποιήθηκαν ως μάρτυρες. Πειραματική Διαδικασία: Τα δείγματα μετά την διαδικασία της ανοσοιστοχημείας μελετήθηκαν με ηλεκτρονικό μικροσκόπιο και η έκφραση τους βαθμολογήθηκε σύμφωνα με το γερμανικό ημι-ποσοτικό σύστημα βαθμολόγησης λαμβάνοντας υπόψιν την ένταση χρώσης και την έκταση (PPSI).Συμπεράματα: Οι τιμές έκφρασης (PPSI) και των 6 υπο μελέτη δεικτών στις προεκλαμπτικές ασθενείς ήταν στατιστικά σημαντικά υψηλότερες σε σύγκριση με την ομάδα ελέγχου (p
