Effect of temperature and Cry1Ab toxin of Bacillus thuringiensis on the biology of Ostrinia nubilalis (Lepidoptera: Crambidae) and Sesamia nonagrioides (Lepidoptera: Noctuidae)

In the present PhD Thesis dissertation we studied the effect of high and low temperatures, as well of the Cry1Ab toxin of Bacillus thuringiensis Berliner regarding the biology of the two most destructive lepidopteran pests of maize (Zea mays L.), Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae) and Sesamia nonagrioides Lefébvre (Lepidoptera: Noctuidae), in Greece. One of our objectives was to search for resistance alleles in natural populations of S. nonagrioides derived from three different geographical locations in Greece and determine the initial frequency of resistance alleles to transgenic corn expressing Cry1Ab insecticidal protein of B. thuringiensis by using the F2 screen on field populations. When resistance alleles are believed to be rare and recessive, the most efficient method is an F2 screen which preserves genetic variation among isofemale lines and concentrates resistance alleles into homozygous genotypes of the F2 generation so that they can be detected. So far, F2 screens have never been developed or used for detecting resistance in S. nonagrioides. In total 75 lines were screened for survival of F2 larvae on Cry1Ab corn leaves. No major resistance alleles were found. The expected frequencies were 7.8 x 10⁻³, 14.7 x 10⁻³ and 7.8 x 10⁻³ for the populations of Thessaloniki, Larissa and Serres respectively. The experiment-wise detection probability of resistance was 94.0%. Evidence of alleles conferring partial resistance to Cry1Ab was found only for the population Thessaloniki. The frequency of alleles for partial resistance was estimated at 6.5 x 10⁻³ with a 95% CI between 8 x 10⁻⁴ and 17.8 x 10⁻³ and a detection probability of 94%. Our results suggest that the frequency of alleles conferring resistance to Cry1Ab, regarding the population of S. nonagrioides, may be rare enough so that the high dose/refuge strategy could be applied with success for resistance management. Susceptibility to Cry1Ab toxin of B. thuringiensis was determined for O. nubilalis larvae collected from two different populations, one of northern Greece (Serres) and one of south-western Germany (Freiburg), and as well for S. nonagrioides larvae collected from three different localities of Greece. First instar larvae of both insects were exposed to artificial diet treated with increasing Cry1Ab concentrations using overlay techniques. Mortalities were evaluated after 7 days. The LC₅₀ (ng/cm²) values of Cry1Ab protein for the two different populations of O. nubilalis did not differ significantly and ranged from 20.2 for the german population to 29.3 for the greek population. Similarly, the LC₅₀ for S. nonagrioides in Greece does not show particular differences in the susceptibility, ranging from 21.3 to 29.2 ng/cm². These results provide an initial baseline for estimating potential shifts in susceptibility that might result from selection and exposure to Cry1Ab-expressing corn hybrids in the future. Moreover, we studied the development of O. nubilalis larvae in various concentrations of Cry1Ab toxin of B. thuringiensis. For this aim we used first instar larvae from two different populations, one of northern Greece (Serres) and one of south-western Germany (Freiburg), which were exposed for 7 days to the same conditions as mentioned previously, and then we determined their larval instar based on the width of their head capsule. Our results show that larval development is significantly affected by the concentration of Cry1Ab toxin, in both populations. As the concentration of Cry1Ab toxin increases, larval development is reduced. Laboratory studies were conducted in order to assess the ability of O. nubilalis to withstand short term exposures to high temperatures. Immature and mature stages of O. nubilalis were used to determine the temperature and time needed to cause mortality in 50% of the population. According to our results, pupal and adult stages were the most tolerant in relation to the larval instars and the egg stage. Among the larval instars, the 4th was the most tolerant while the 1st appeared to be the most susceptible. Overall no differences were observed between males and females regarding the pupal and adult stage. Regarding the susceptibility to the toxin Cry1Ab of B. thuringiensis of O. nubilalis larvae that originated from adults that were exposed to high temperatures, increasing exposure temperature of adults from 43°C to 44°C or even to 45°C resulted in enhanced susceptibility of O. nubilalis larvae to the toxin Cry1Ab of B. thuringiensis. On the contrary, elevating exposure time from 15 min to 30 min or even 60 min does not appear to influence considerably susceptibility of O. nubilalis larvae to the toxin Cry1Ab of B. thuringiensis, as no significant differences were observed among the time intervals. The age-dependent cold hardiness profile of O. nubilalis is compared between non-diapausing and diapausing larvae, as well as with field-collected larvae. The results suggest that both cold tolerance and accumulation of cryoprotectants depends on the age of O. nubilalis larva. Late fifth-instar non-diapausing larvae are more cold tolerant than younger larvae, as they show enhanced ability to withstand sub-zero temperatures. No appreciable difference is observed between the experimental groups of diapausing larvae as far as their supercooling ability and tolerance at sub-zero temperatures above the supercooling point is concerned. In general, both field-collected and diapausing larvae are more cold tolerant than non-diapausing larvae, indicating a direct link between diapause and cold hardiness. The age of diapausing larvae affects the ability to accumulate glycerol. Glycerol levels of 45-day old diapausing larvae are significantly higher (2.7 fold) compared with 90-day old diapausing larvae. Moreover, diapausing larvae display a 5-13 fold higher glycerol content compared with non-diapausing larvae. Finally, we studied the cold hardiness profile of S. nonagrioides among non-diapausing and diapausing laboratory larvae, as well as with field-collected larvae, taking into consideration various parameters, such as supercooling ability, mean lethal temperature and accumulation of cryoprotectant substances, in relation to diapause. Our results show that S. nonagrioides not only displays a small ability of supercooling but at the same time we did not observe significant difference between diapausing and non diapausing larvae. Moreover, no significant mortality was achieved in temperatures higher than their mean supercooling point, while most of the individuals died at temperatures slightly lower than their mean supercooling point after extended exposure to them. Regarding the role of cryoprotectant substances, accumulation of glycerol seems to be closely related to diapause, in contrast to accumulation of trehalose, which is more related to low temperatures higher than 0°C. However, a direct link between cold hardiness and accumulation of both glycerol and trehalose in the tissues of the larvae was found.Στην παρούσα διδακτορική διατριβή μελετήθηκαν ορισμένα στοιχεία της βιολογίας των δύο κυριότερων και πιο επιβλαβών Λεπιδόπτερων εντομών της καλλιέργειας του καλαμποκιού του Ostrinia nubilalis (Hubner) (Lepidoptera Crambidae) και Sesamia nonagrioides Lefebvre (Lepidoptera Noctuidae) σε σχέση με τη θερμοκρασία και την τοξίνη Cry1Ab του Bacillus thuringiensis Berliner. Συγκεκριμένα μελετήθηκε η πιθανότητα εύρεσης γονίδιων ανθεκτικότητας του S. nonagrioides στην τοξίνη Cry1Ab του B. Thuringiensis. Για το σκοπό αυτό χρησιμοποιήθηκαν 3 διαφορετικοί πληθυσμοί αγρού του S. nonagrioides (Θεσσαλονίκη, Λάρισα και Σέρρες). Η αναμενόμενη συχνότητα γονιδίων ανθεκτικότητας E[PR] ήταν 7.8 x 10⁻³, 14.7 x 10⁻³ και 7.8 x 10⁻³ για τους πληθυσμούς από τη Θεσσαλονίκη, τη Λάρισα και τις Σέρρες αντίστοιχα. Τα αποτελέσματα μας δείχνουν ότι η πιθανότητα εύρεσης γονιδίων ανθεκτικότητας του εντόμου S. nonagrioides στην τοξίνη Cry1Ab είναι μικρή έτσι ώστε τελικά η στρατηγική της «υψηλής δόσης-καταφυγίου» να μπορεί να εφαρμοστεί με επιτυχία σε ένα πρόγραμμα διαχείρισης ανθεκτικότητας σε γενετικά τροποποιημένη καλλιέργεια καλαμποκιού που εκφράζει την τοξίνη Cry1Ab του Β. thuringiensis. Επίσης μελετήθηκε η επίδραση της τοξίνης Cry1Ab του Β. thuringiensis στην επιβίωση των προνυμφών του Ο. nubilalis και του S. Nonagrioides. Τόσο στο Ο. nubilalis όσο και στο S. nonagrioides δεν παρατηρήθηκαν σημαντικές διαφορές μεταξύ των διαφορετικών πληθυσμών ως προς την ευαισθησία τους στην τοξίνη Cry1Ab του Β. thuringiensis. Επιπλέον μελετήθηκε και η ανάπτυξη των προνυμφών του Ο. nubilalis σε διάφορες συγκεντρώσεις της τοξίνης Cry1Ab του Β. thuringiensis. Από τα αποτελέσματα προκύπτει ότι υπάρχει σημαντική διαφορά στην ανάπτυξη των προνυμφών όταν αυτές εκτεθούν σε διάφορες συγκεντρώσεις της τοξίνης Cry1Ab του Β. thuringiensis και στους δύο πληθυσμούς. Όσο αυξάνεται η συγκέντρωση της τοξίνης τόσο επιβραδύνεται η ανάπτυξη των προνυμφών. Η αντοχή του Ο. nubilalis σε βραχείας διάρκειας έκθεση σε υψηλές θερμοκρασίες μελετήθηκε εργαστηριακά. Τα πιο ανθεκτικά στάδια ήταν αυτά του ενήλικου και της νύμφης. Σε καμία από τις παραπάνω μεταχειρίσεις δεν παρατηρήθηκε σημαντική διάφορα μεταξύ αρσενικών και θηλυκών ατόμων. Το πιο ευαίσθητο στάδιο ήταν αυτό του αβγού. Όσον άφορα στη μελέτη της ευαισθησίας προνυμφών του Ο. nubilalis στην τοξίνη Cry1Ab του Β. thuringiensis που προήλθαν από ενήλικα που δέχθηκαν την επίδραση υψηλών θερμοκρασιών αύξηση της θερμοκρασίας έκθεσης των ενήλικων πέραν τους 43°C είχε ως αποτέλεσμα την αύξηση της ευαισθησίας των προνυμφών που προέκυψαν από τα ενήλικα αυτά στην τοξίνη Cry1Ab του Β. thuringiensis. Η επίδραση της ηλικίας των προνυμφών του Ο. nubilalis στην αντοχή στο ψύχος και τη συσσώρευση των κρυοπροστατευτικών ουσιών εξετάσθηκε με μια σειρά πειραμάτων. Οι 7 ημερών μη διαπαύουσες προνύμφες 5ης ηλικίας ήταν περισσότερο ανθεκτικές στο ψύχος σε σύγκριση με τις μικρότερης ηλικίας μη διαπαύουσες προνύμφες. Αντίθετα στις διαπαύουσες προνύμφες η ηλικία δεν παίζει σημαντικό ρόλο. Ωστόσο η ηλικία των διαπαυουσών προνυμφών φαίνεται να επηρεάζει σημαντικά την ικανότητα συσσώρευσης της γλυκερόλης καθώς διαπαύουσες προνύμφες ηλικίας 45 ήμερων παρουσίασαν κατά 2.7 φορές μεγαλύτερη συγκέντρωση γλυκερόλης έναντι των διαπαυουσών προνυμφών ηλικίας 90 ημερών. Επιπλέον οι διαπαύουσες προνύμφες είχαν 5-13 φορές υψηλότερη συγκέντρωση γλυκερόλης σε σύγκριση με τις μη διαπαύουσες προνύμφες. Τέλος μελετήθηκε η αντοχή σε χαμηλές θερμοκρασίες του S. nonagrioides. Από τα αποτελέσματα προκύπτει ότι το S. nonagrioides όχι μόνο εμφανίζει μικρή σχετική ικανότητα υπέρψυξης αλλά παράλληλα δεν παρατηρήθηκαν σημαντικές διαφορές μεταξύ διαπαυουσών και μη διαπαυουσών προνυμφών. Όσον αφορά στο ρόλο των κρυοπροστατευτικών ουσιών αν και σε γενικές γραμμές δεν παρατηρήθηκε συσσώρευση μεγάλων ποσοτήτων κρυοπροστατευτικών ουσιών εντούτοις βρέθηκε η συσσώρευση της γλυκερόλης να σχετίζεται άμεσα με τη διάπαυση σε αντίθεση με την συσσώρευση της τρεχαλόζης η οποία σχετίζεται περισσότερο με την έκθεση σε χαμηλές θερμοκρασίες μεγαλύτερες από τους 0°C
