Study of lipids focusing on molecular structures of phosphonolipids from edible molluscs (mussels, snails) and knidozoa (medusae)

The mam purpose of the present dissertation was to study the existence and the structure of molecular species of natural phosphonolipids. Their accurate role m nature is not yet well understood and this fact arouses the scientific interest in the investigation of their biological significance. Moreover, phosphonolipids due to the stability of their C-P bonds, have been found also in human tissues where as in the case of other mammals, there is evidence that phosphonolipids are not biosynthetic products but they are taken up from food and incorporated into intrinsic lipids and therefore we have to elucidate how phosphonolipids affect human health. So, we decided to perform our experiments on organisms (like molluscs and cnidaria) that are a rich source of phosphonolipids according to references, in order to study the variety of their molecular structures, having as final purpose the contribution to the investigation of their functional roles. More specifically, the mussel M. galloprovincialis and the land snail E. vermiculaia were selected from the molluscs and the medusae A. aurita and P. noctiluca from the cnidaria. The first two of them are common food for human and therefore further research to these molecules is additionally justified. It is pointed out that the study on the mussel and on the medusa A. aurita was extended to the analysis of all the other phospholipid classes, since such a study had not been performed before.The theoretical part of the dissertation includes a literature review on natural phosphonic acids and phosphonolipids where the significance and their possible physiological role are also mentioned. In addition, several applications of the synthetic phosphonic derivatives are reported. Moreover, related data on the examined organisms and their lipids are given. The results of this study are summarized below.EDIBLE MOLLUSCS: Total lipids of the mussel M. galloprovincialis, were extracted from the whole body (1,27% of the fresh tissue) and were fractionated into polar (61,5%) and neutral (38,5%) lipid classes. The phosphonolipids were found to constitute 14% of mussel phospholipids. They contain one ceramide aminoethylphosphonate species (11,2%) and its N-methyl analog (2,8 %); the latter is more polar due to the presence of a higher percentage of unsaturated fatty acids (+23,53%) and hydroxy fatty acids (+12,93%) as well as due to the presence of a higher percentage of trihydroxy sphin- gosine bases (+34,5%).The rest phospholipids of the mussel were separated into: phosphatidylcholine 41,6 % (of which 11,3 % corresponds to glyceryl ethers); phosphatidylethanolamme, which consists of 2 molecular analogs PE], 13,5% (24% glyceryl ethers) and PE2, 13,1% ; phosphatidylinositol, 6,7% ; lyso - phosphatidylcholine, 1,6 % and the remaining 4,5 % was distributed to glycophospholipids.The saturated fatty acids are predominant in all two lipid classes (polar and neutral lipids) of the mussel in contrast to the mussels of other geographical regions where the unsaturated fatty acids are predominant In the case of the land snail E. vermiculata, sphingophosphonohpid fraction was found to constitute 9,0% of the phospholipids and consisted of the already reported in the literature ceramide aminoethyl- phosphonate species, 7,5 %, and the ceramide methylaminoethyl phosphonate species identified in the present dissertation (1,5%); the latter is more polar due to the presence of hydroxy fatty acids (+37,8%) and trihydroxy sphingosme bases (+36,25%).Unsaturated fatty acids are predominant in all two lipid classes (polar and neutral lipids).MEDUSAE : Total lipids of the medusa A. aurita. were extracted from the whole body (0,04 % of the fresh tissue) and were fractionated into neutral (64,2 %) and polar (35,8 %) lipid classes.The phosphonolipids were found to be 21,7 % of^espU^Jrand consist of a ceramide ammoethylphos- phonate species (18,3 %) and 3 N-methyl analogs m percentages of 1,3 %, 1,1 % and 1,0 % respectively, m order of increasing polarity. The first N-methyl analog was more polar due to the increased percentage of the unsaturated fatty acids (+16,01 %) and the trihydroxy sphingosine bases (+6,17 %), the second N-methyl analog for the same reasons (+9,02% and +16,88%, respectively) and the third one due to the higher percentage of hydroxy fatty acids (10,37%) and trihydroxy sphingosine bases (+13,26%).The rest phospholipids of A. aurita were separated into: phosphatidylcholine 43,1 % (of which 36,2 % corresponds to glyceryl ethers), phosphatidylethanolamine 19,1 % (4.5 % glyceryl ethers), cardiolipin5,2 %, phosphatidylinositol 2,6 % and lyso-phosphatidylcholine 5,0 %.Saturated fatty acids are predominant in all two lipid classes (polar and neutral lipids).In the case of the medusa P. noctiluca, phosphonolipids were found to be 24,3 % of phospholipids and consist of the already reported in the literature ceramide aminoethylphosphonate species, 21%, and two N-methyl analogs in percentages of 2,0% and 1,3% respectively, in order of increasing polarity. The first N-methyl analog was more polar due to the increased percentage of the unsaturated fatty acids (+18,41%) and trihydroxy sphingosine bases (+48.49%) and the second one due to a higher percentage of hydroxy fatty acids (+8,49%) and trihydroxy sphingosine bases (+12.29%).In this work, a variability of ceramide moiety occurred in the molecular species of ceramide aminoethylphosphonate and their N-methyl analogs has been observed, because of the presence of a higher percentage of hydroxy fatty acids and the presence of an increasing percentage of trihydroxy sphingosine bases. So, there is an increase by one or two hydroxyl groups in these molecules, causing the increase of their stability. The inert character of phosphonolipids may be used to generate membranes or structural components of organisms, which are resistant to hydrolysis, oxidation or other metabolic processes. This is in agreement with the fact that especially in the medusae, which, as it is known, are extremely resistant to polluted environment, a great number of such molecular structures were identified.The variety of the ceramide aminoalkylphosphonate structures, identified in the present study, referred to the whole body of the examined organisms. It would be of further interest to determine if the presence of ceramide aminoalkylphosphonate species is localized in specific tissues of the examined molluscs or medusae. It is known that in the manne invertebrates (gastropod molluscs), the ceramide phosphonate species are distributed to various tissue types, like the nervous (ganglion and fibers) and other tissues (skin, viscera, muscle tissue). The biological significance of these distnbutions is not clarified, but they may suggest differences in the metabolism of these molecules between the tissues. If such a localization exists it might contnbute to understanding the functional role of phosphonolip- lds.Moreover, the differences in the phosphonolipid structure and content between the various tissues are interesting in phylogenetic studies as well as in studies of the food chain.Finally, it has been already recognized that the C-P compounds show antibacterial, antifungal and antiviral effects and in addition the sphmgosines and the cerarrudes present biological activity. So it would be interesting to investigate if the natural phosphonolipids can be used as chemotherapeutic agents in the future. Thus, the study of organisms that are rich in phosphonolipids, and the investigation of their lipid molecular diversity, are of interest.Στην παρούσα εργασία μελετήθηκε η δομή μοριακών ειδών φυσικών φωσφονολιποειδών των οποίων ο ρόλος στη φύση είναι ακόμη αδιευκρίνιστος και διεγείρει το ενδιαφέρον των επιστημόνων ως προς τη διερεύνηση της σημασίας τους. Επιπλέον, τα φωσφονολιποειδή ως αδιάσπαστα μόρια ενσωματώνονται στα λιποειδή των μεμβρανών του ανθρώπου μέσα από την τροφική αλυσίδα και απαιτεί απάντηση το ερώτημα πως εν τέλει επηρεάζουν την υγεία του. Έτσι σαν πειραματικό αντικείμενο της μελέτης μας επελέγησαν οργανισμοί που αποτελούν πλούσια πηγή φωσφονολιποειδών (όπως μαλάκια και κνιδάρια) προκειμένου να μελετηθεί η ποικιλία των μοριακών δομών τους με απώτερο στόχο τη συμβολή στην διερεύνηση του ρόλου τους. Συγκεκριμένα επελέγησαν από τα μαλάκια το μύδι Μ. galloprovincialis και το σαλιγκάρι E. vermiculata και από τα κνιδάρια οι μέδουσες A. attrita και Ρ. noctiluca. Τα 2 πρώτα αποτελούν επιπλέον ευρείας κατανάλωσης τρόφιμα για τον άνθρωπο. Επιση- μαίνεται ότι στο μύδι και στην μέδουσα A. aurita επεξετάθη η έρευνα και στην ανάλυση όλων των υπόλοιπων τάξεων των φωσφολιποειδών τους αφού δεν είχε γίνει προηγούμενα τέτοια μελέτη.Στο θεωρητικό μέρος της εργασίας γίνεται βιβλιογραφική ανασκόπηση για τα φυσικά φωσφονικά οξέα και τα φωσφονολιποειδή όπου αναφέρονται οι πιθανοί φυσιολογικοί τους ρόλοι. Ακόμη, αναφέ- ρονται οι εφαρμογές των συνθετικών φωσφονικών παραγώγων. Επίσης δίνονται στοιχεία που αφορούν τους εξεταζόμενους οργανισμούς και τα λιποειδή τους. Τα αποτελέσματα της εργασίας αυτής είναι τα παρακάτω :ΒΡΩΣΙΜΑ ΜΑΛΑΚΙΑ : Ως προς το μύδι Μ. galloprovincialis, τα ολικά λιποειδή εκχυλίστηκαν από ολόκληρο το σώμα (1,27% του νωπού ιστού) και κλασματώθηκαν σε πολικά 61,5 % και ουδέτερα 38,5 % Το 14 % των φωσφολιποειδών του μυδιού ανήκε σε φωσφονολιποειδή που αποτελούντο από ένα κη- ραμιδυλοαμινοαιθυλοφωσφονικό οξύ (11,2 %) και το Ν-μέθυλο ανάλογο του (2,8 %) το οποίο είναι πολικότερο λόγω μεγαλύτερου ποσοστού ακόρεστων λιπαρών οξέων (+23,53 %) καιυδρόξυ λιπαρών οξέων (+12,93 %) καθώς και μεγαλύτερου ποσοστού τριυδρόξυ σφιγγοσινούχων βάσεων (+34,5 %). Τα υπόλοιπα φωσφολιποειδή του μυδιού κλασματώθηκαν στα : φωσφατιδυλοχολίνη, 41,6 % (11,3 % ανήκει σε γλυκεριναιθέρες), φωσφατιδυλοαιθανολαμίνη αποτελούμενη από 2 μοριακά ανάλογα ΡΕι, 13,5 % (24 % γλυκεριναιθέρες) και ΡΕ2, 13,1 %, φωσφατιδυλοινοσιτόλη, 6,7 %, λυσο - φωσφατιδυ- λοχολίνη, 1,6 % και το υπόλοιπο 4,5 % κατανεμήθηκε σε γλυκοφωσφολιποειδή.Κορεσμένα λιπαρά οξέα επικρατούν και στις 2 τάξεις λιποειδών (πολικά και ουδέτερα) του μυδιού εν αντιθέσει με τα μύδια άλλων γεωγραφικών περιοχών όπου υπερέχουν τα ακόρεστα.Ως προς το σαλιγκάρι E. vermiculata, το 9 % των φωσφολιποειδών ανήκει σε σφιγγοφωσφονολι- ποειδή που αποτελούντο από το ήδη γνωστό στην βιβλιογραφία, κηραμιδυλοαμινοαιθυλοφωσφονικό οξύ, 7,5 % και από το ταυτοποιηθέν στην παρούσα εργασία Ν-μέθυλο ανάλογο του (1,5 %) το οποίο είναι πολικότερο λόγω παρουσίας σε αυτό υδρόξυ λιπαρών οξέων (+37,8%) και τριυδρόξυ σφιγγοσινούχων βάσεων (+36,25 %).Ακόρεστα λιπαρά οξέα επικρατούν και στις 2 τάξεις λιποειδών του (πολικά και ουδέτερα). ΜΕΔΟΥΣΕΣ : Ως προς τη μέδουσα A. aurita, τα ολικά λιποειδή εκχυλίστηκαν από ολόκληρο το σώμα (0,04 % του νωπού ιστού) και κλασματώθηκαν σε ουδέτερα 64,2 % και πολικά 35,8 %.Το 21,7 % των φωσφολιποειδών της Λ. aurita ανήκε σε φωσφονολιποειδή που αποτελούντο από ένα κηραμιδυλοαμινοαιθυλοφωσφονικό οξύ (18,3 %) και από 3 Ν-μέθυλο ανάλογα του με ποσοστά 1,3 %, 1,1 % και 1,0 %, αντίστοιχα κατά σειρά αυξανόμενης πολικότητας. Το πρώτο Ν-μέθυλο ανάλογο είναι πολικότερο λόγω αύξησης του ποσοστού των ακόρεστων λιπαρών οξέων (+16,01 %) και των τριυδρόξυ σφιγγοσινούχων βάσεων (+6,17 %), το δεύτερο ανάλογο για τους ίδιους λόγους (+9,02 % και +16,88 %, αντίστοιχα) και το τρίτο λόγω περισσότερων υδρόξυ λιπαρών οξέων (10,37 %) και τριυδρόξυ σφιγγοσινούχων βάσεων (+13,26 %).Τα υπόλοιπα φωσφολιποειδή της A. aurita κλασματώθηκαν στα : φωσφατιδυλοχολίνη 43,1 % (36,2 % ανήκε σε γλυκεριναιθέρες), φωσφατιδυλοαιθανολαμίνη 19,1 % (4,5 % γλυκεριναιθέρες), καρδιολι- πίνη 5,2 %, φωσφατιδυλοινοσιτόλη 2,6 % και λυσο-φωσφατιδυλοχολίνη 5,0 %.Κορεσμένα λιπαρά οξέα επικρατούν και στις 2 τάξεις λιποειδών της (πολικά και ουδέτερα).Ως προς τη μέδουσα/’, noctiluca, το 24,3 % των φωσφολιποειδών ανήκε σε φωσφονολιποειδή που απαρτίζοντο από το ήδη γνωστό στην βιβλιογραφία, κηραμιδυλοαμινοαιθυλοφωσφονικό οξύ, 21 % και από τα δύο Ν-μέθυλο ανάλογα του με ποσοστά 2,0 % και 1,3 %, αντίστοιχα κατά σειρά αυξανόμενης πολικότητας. Το πρώτο Ν-μέθυλο ανάλογο είναι πολικότερο λόγω αύξησης του ποσοστού ακόρεστων λιπαρών οξέων (+18,41 %) και τριυδρόξυ σφιγγοσινούχων βάσεων (+48,49 %) και το δεύτερο Ν-μέθυλο ανάλογο λόγω μεγαλύτερου ποσοστού υδρόξυ λιπαρών οξέων (+8,49 %) και τριυδρόξυ σφιγγοσινούχων βάσεων (+12,29 %).Παρατηρήθηκε στην παρούσα εργασία ότι στα μοριακά είδη του κηραμιδυλοαμινοαιθυλοφωσφο- νικού οξέος και στα Ν-μέθυλο ανάλογα τους, που ταυτοποιήθηκαν ποιοτικά και ποσοτικά, εμφανίζονται διαφορές στα κηραμίδια που οφείλονται στην παρουσία μεγαλύτερου ποσοστού υδρόξυ λιπαρών οξέων και στην παρουσία αυξανόμενου ποσοστού τριυδρόξυ σφιγγοσινούχων βάσεων. Δηλαδή υπάρχει αύξηση κατά μία ή δύο υδροξυλομάδες ανά δύο μόρια, γεγονός που αυξάνει τον αδρανή χαρακτήρα του μορίου. Τέτοια μόρια είναι δυνατόν να χρησιμοποιηθούν για να δημισυργηθούν μεμβράνες ή δομικά συστατικά οργανισμών ανθεκτικά σε υδρολύσεις, οξειδώσεις ή άλλες μεταβολικές πορείες. Η παραπάνω αναφορά είναι σε συμφωνία με το γεγονός ότι ειδικά στις μέδουσες οι οποίες ως γνωστόν είναι εξαιρετικά ανθεκτικές σε δυσμενείς συνθήκες περιβάλλοντος, ταυτοποιήθηκε μεγάλος αριθμός τέτοιων μοριακών δομών.Η ποικιλία των δομών των φωσφονολιποειδών που ταυτοποιήθηκαν στην παρούσα εργασία αφορούσε ολόκληρο το σώμα των εξεταζόμενων οργανισμών. Θα είχε πρόσθετο ενδιαφέρον να διερευνη- θεί η παρουσία εκάστου μοριακού είδους σε ποιούς συγκεκριμένους ιστούς των εξεταζόμενων μαλακίων ή μεδουσών αντιστοιχούν. Είναι γνωστό ότι στα θαλάσσια ασπόνδυλα (γαστερόποδα μαλάκια) τα φωσφονολιποειδή κατανέμονται σε διάφορα είδη ιστών όπως στους νευρικούς (γάγγλια και ίνες) και σε άλλους ιστούς (δέρμα, σπλάχνα, μυϊκός ιστός) Η βιολογική σημασία των κατανομών αυτών δεν έχει διευκρινιστεί αλλά μία άποψη είναι ότι οι διαφορές τους δυνατόν να οφείλονται σε διαφορές στον μεταβολισμό των μορίων αυτών στο συγκεκριμένο ιστό. Έτσι, ένας τέτοιος εντοπισμός θα συνέβαλλε στη διερεύνηση του ρόλου των φωσφονολιποειδών.Επίσης οι διαφορές στη σύσταση και στην περιεκτικότητα των φωσφονολιποειδών μεταξύ των διαφόρων ιστών θα συνέβαλε περαιτέρω στην διερεύνηση της τροφικής αλυσίδας.Τέλος, έχει αναγνωρισθεί ότι οι C-P ενώσεις έχουν αντιβακτηριακή αντιμυκητιακή και αντιιική δράση και επιπλέον οι σφιγγοσίνες και τα κηραμίδια παρουσιάζουν βιολογική δραστικότητα. Κατόπιν αυτού έχει σημασία να εξετασθεί αν τα φυσικά φωσφονολιποειδή έχουν τη δυνατότητα να συμβάλλουν στη χημειοθεραπευτική σαν πρωτογενείς πηγές δραστικών ουσιών για την παρασκευή μελλοντικών φαρμάκων. Συνεπώς έχει ενδιαφέρον ο εντοπισμός των σχετικών οργανισμών που είναι πλούσια και φθηνή πηγή φωσφονολιποειδών και η μελέτη της ποικιλίας των μοριακών δομών σε αυτά
