Taxonomic and economical study on the Agaricales of Greece - contribution of the pure cultures

The present dissertation is aiming at the study of species of Agaricales (Basidiomycetes) from a taxonomic and ecological point of view, with emphasis on the contribution of the characteristics and properties they exhibit in pure culture.The fungal biodiversity, particularly in Greece, is inadequately studied and the available data are limited. The recorded species of Basidiomycetes from Greece correspond to less than half of the overall recorded fungal species, and most records constitute simple references, lacking any descriptions or taxonomical comments. In addition, dried specimens have not been kept for most of these species, to serve as reference material. Finally, the data concerning the characteristics and properties exhibited in pure culture are almost inexistent.Moreover, this group of Basidiomycetes includes a large number of genera and species with significant taxonomical and nomenclatural problems. This is due to the fact that their taxonomy has, until recently, been based solely on morphological characteristics of the basidiocarps, that are very variable in respect to the environmental conditions or the different stages of maturity of the basidiocarps. The study of the morphological characteristics and the various properties of the mycelium of these species when isolated in pure culture has proved to contribute significant data that, in combination with physiological, biochemical, genetic and molecular data, can contribute to the better understanding of the relationships between the various taxa, and, in effect, the creation of a natural classification system.The present study deals with material from species that have traditionally been placed in the order Agaricales, and are characterized by fleshy, ephemerous basidiocarps with a lamellate hymenophore. According to modern classification systems, these species belong to more than four orders.The specimens have been collected from various habitats, mostly from forests, and from various locations, mainly in Central and Southern Greece. Collections were made mostly in autumn and winter and rarely spring. An effort has been made to isolate the collected specimens in pure culture. The material included in the present study is a small part of the overall collected material and is restricted to genera that have been isolated and studied in pure culture. The species are kept in dried state in the Mycotheca (Mycological Herbarium) of the University of Athens and the cultured strains isolated from these species in the Culture Collection of Fungi ATHUM of the University of Athens. The macroscopical and microscopical characteristics of 235 specimens of basidiocarps have been studied and 57 taxa in total have been determined (53 species and 4 varieties), belonging to 16 genera, 9 families and 4 orders. 89 of these specimens have been isolated in pure culture, corresponding to 31 taxa (29 species and 2 varieties) that belong to 15 genera, 9 families and 4 orders. The colonies of these strains have been studied macroscopically and microscopically and the strains where traced for the enzymes laccase and tyrosinase with spot tests and specific reagents.In the above-mentioned specimens, they are included specimens and their isolates that have been determined at the generic level and most possibly belong to two new taxa.The present study includes 18 taxa that are newly recorded for Greece, whereas 13 taxa are recorded for the second or third time. One variety is newly recorded for Europe. 33 taxa are recorded in new habitats or substrates for Greece, including the newly recorded taxa. Of these records, one is new for Europe and one at a global level. Taxa that are newly recorded from forests of Abies cephalonica are also new worldwide records for that specific habitat.According to the World Directory of Culture Collections, published by the World Data Center for Microorganisms (WDCM, 2003), the strains of six species are unique globally, whereas four more strains are kept in one or two other Culture Collections.The macroscopical and microscopical study of the collected material resulted to the following taxonomical remarks:There exists in some cases a significant variation and fluctuation in the macroscopical and microscopical characteristics between individual specimens of the same collection or between different collections of the same species, as, for example, in species of the genera Lentinellus and Rhodocybe.In some species, such as Clitopilus prunulus and Rhodocybe gemina, there are significant divergences from descriptions in the literature, of which the most important concern the size and morphology of the basidiospores, as well as the morphology of the cystidia or the pileipellis.Microscopical characteristics have been observed in some species that have not been previously reported. Such is the case in species of genera Amanita and Limacella, in which the presence of thick-walled basidia and basidiospores is striking. This characteristic is possibly an adaptation to environmental conditions.A significant number of species belong to groups of closely related species with a similar or partly overlapping range in their morphological characteristics, thus making the limits for their determination unclear and the further comparative study with other methods imperative. Such examples exist in various groups from different genera, such as group Vaginatae in genus Amanita, Dapetes in Lactarius and Omphalodei in Lentinellus.The habitat or substrate from which a species has been collected may often be considered characteristic, but this may not always be a safe supplementary criterion for the determination or separation of closely related species. For example, the distinction of closely related species Lactarius deliciosus and L. salmonicolor on the basis of their appearance in forests of Pinus spp. and Abies spp. respectively, is considered precarious. Species such as Amanita phalloides, Lactarius chrysorrheus, Lactarius aurantiacus and Hohenbuehelia auriscalpium appear, according to the literature, in deciduous forests, the former three usually and the latter exclusively so, whereas in the present study they have been found in pure forests of Abies cephalonica.Moreover, the study of the characteristics of the isolates in pure culture has resulted in the following deductions:The formation of anamorphs and teleomorphs on a hemi-synthetic substrate in an unusual phenomenon, and, when they do appear, they are considered important taxonomic features. Anamorphs, such as conidia, chlamydospores and sclerotia, and teleomorphs have been observed in 16 taxa. In 10 of these taxa, they are newly recorded globally.The macroscopical and microscopical characteristics of colonies in pure culture can differ significantly between species of the same genus, as in Lentinellus, or they may exhibit some important similarities that may characterize a genus, as in Hohenbuehelia and Pleurotus. In some cases, they may lead to groupings of species that correspond to intrageneric taxa, as in Amanita. In certain colonies, some features are so characteristic and constant that a safe determination of the species or the genus can be ascertained without the presence of basidiocarps. Finally, these characteristics when viewed comparatively occasionally exhibit certain similarities at the generic or a higher level that suggest possible reationships between them.Some microscopical characteristics of the basidiocarps, such as the presence of clamps or of modified hyphae, are constant and appear also in isolates in pure culture.In many strains the production of usually coloured metabolites is evident. The metabolites may diffuse in the substrate or they may be secreted as droplets on the surface of the colony. The production or the quantity of the produced metabolites often seems to be related to the production of basidiocarps or sclerotia, as well as with states of stress, such as unfavorable growth conditions or competition.A significant number of strains has been found to produce laccase, an indication of ligninolytic capability. Tyrosinase was traced in relatively few strains. The production of these enzymes seems to be correlated with certain taxa, whereas their activity seem to be dependent on the strain.The results of the present study enrich the data concerning the diversity of Basidiomycetes in Greece and contribute to the better knowledge of the various taxa and to the solution of the taxonomical problems in the Agaricales.Αντικείμενο της παρούσας διατριβής αποτελεί η μελέτη ειδών των Agaricales (Βασιδομυκήτων) από ταξινομική και οικολογική άποψη, με έμψαση στη συμβολή των γνωρισμάτων και των ιδιοτήτων που εμφανίζουν κατά την απομόνωσή τους σε καθαρές καλλιέργειες.Η βιοποικιλότητα των μυκήτων ειδικά στην Ελλάδα είναι ανεπαρκώς μελετημένη και τα δεδομένα αρκετά περιορισμένα, γεγονός που απετέλεσε έναν από τους βασικούς λόγους για την πραγματοποίηση της συγκεκριμένης έρευνας. Οι αναφορές ειδών Βασιδιομυκήτων αντιστοιχούν σε αριθμό μικρότερο του μισού του συνολικού αριθμού ειδών μυκήτων και οι περισσότερες αποτελούν απλές καταγραφές, χωρίς να δίνονται περιγραφές ή ταξινομικά σχόλια. Επιπλέον, για τα περισσότερα από τα είδη αυτά δεν υπάρχει αποξηραμένο δείγμα ερμπαρίου, που να φυλάσσεται ως υλικό αναφοράς. Παράλληλα, τα δεδομένα, όσον αφορά στα χαρακτηριστικά και τις ιδιότητές τους σε καθαρές καλλιέργειες, είναι σχεδόν ανύπαρκτα.Επιπροσθέτως, στην ομάδα αυτή των Βασιδιομυκήτων υπάγεται ένας μεγάλος αριθμός γενών και ειδών καθώς και των συνωνύμων τους, με πολλά ταξινομικά και ονοματολογικά προβλήματα. Το γεγονός αυτό αποτελεί απόρροια της ταξινόμησης τους, έως σχετικά πρόσφατα, με βάση αποκλειστικά τους μορφολογικούς χαρακτήρες των βασιδιοκαρπίων, οι οποίοι εμφανίζουν έντονη παραλλακτικότητα, λόγω της άμεσης επίδρασης των περιβαλλοντικών συνθηκών και ποικιλομορφία κατά τα διάφορα στάδια εξέλιξής τους. Η μελέτη των μορφολογικών χαρακτήρων και διαφόρων ιδιοτήτων του μυκηλίου των ειδών αυτών σε καθαρές καλλιέργειες έχει αποδειχθεί ότι μπορεί να συνεισφέρει σημαντικά δεδομένα και, παράλληλα με άλλα, όπως φυσιολογικά, βιοχημικά, γενετικά και μοριακά, μπορεί να βοηθήσει στην ταξινόμηση και στην καλύτερη κατανόηση των σχέσεων ανάμεσα στις διάφορες ταξινομικές μονάδες, για τη δημιουργία ενός φυσικού συστήματος κατάταξης.Στην παρούσα έρευνα μελετήθηκε υλικό από είδη που κατατάσσονται παραδοσιακά στην τάξη Agaricales και χαρακτηρίζονται από σαρκώδη, εφήμερα βασιδιοκάρπια, με υμενοφόρο με μορφή ελασμάτων. Σύμφωνα με σύγχρονα ταξινομικά συστήματα τα είδη αυτά ανήκουν σε περισσότερες από τέσσερις τάξεις.Δείγματα συλλέχθηκαν από ποικίλους βιοτόπους, κυρίως δασικούς, από διάφορες περιοχές κυρίως στην Κεντρική και Νότια Ελλάδα κατά τους φθινοπωρινούς και χειμερινούς μήνες, σπανιότατα τους εαρινούς και έγινε προσπάθεια απομόνωσής τους σε καθαρή καλλιέργεια. Το υλικό που περιλαμβάνεται στην παρούσα μελέτη αποτελεί μέρος του συνόλου των συλλογών και περιορίζεται σε γένη που έχουν μελετηθεί και σε καθαρή καλλιέργεια. Τα δείγματα μετά την επεξεργασία τους φυλάσσονται αποξηραμένα στη Μυκητοθήκη (Μυκητολογικό Ερμπάριο) του Πανεπιστημίου Αθηνών και τα στελέχη των καλλιεργειών, που απομονώθηκαν από αυτά, στη Συλλογή Καλλιεργειών Μυκήτων ATHUM του Πανεπιστημίου Αθηνών.Μελετήθηκαν τα μακροσκοπικά και μικροσκοπικά χαρακτηριστικά των βασιδιοκαρπίων από 235 δείγματα και προσδιορίστηκαν συνολικά 57 ταξινομικές μονάδες (53 είδη και 4 ποικιλίες), που ανήκουν σε 16 γένη, 9 οικογένειες και 4 τάξεις. Παράλληλα, από τα δείγματα αυτά 89 απομονώθηκαν σε καθαρή καλλιέργεια, που αντιστοιχούν σε 31 ταξινομικές μονάδες (29 είδη και 2 ποικιλίες) και ανήκουν σε 15 γένη, 9 οικογένειες και 4 τάξεις. Στα στελέχη αυτά έγινε μελέτη των μακροσκοπικών και μικροσκοπικών χαρακτηριστικών των αποικιών και ανίχνευση των ενζύμων λακκάση και τυροσινάση με σημειακές δοκιμασίες (spot tests) με ειδικά αντιδραστήρια.Στα παραπάνω περιλαμβάνονται δείγματα και απομονώσεις τους, που έχουν προσδιορισθεί σε επίπεδο γένους και τα οποία πιθανότατα ανήκουν σε δύο νέες για την επιστήμη ταξινομικές μονάδες.Στην παρούσα μελέτη περιλαμβάνονται 18 ταξινομικές μονάδες που αποτελούν νέες αναφορές για την Ελλάδα, ενώ 13 ταξινομικές μονάδες αποτελούν δεύτερη έως τρίτη αναφορά. Μία από τις ποικιλίες αναφέρεται για πρώτη φορά στην Ευρώπη.Παράλληλα, 33 ταξινομικές μονάδες καταγράφονται σε νέα ενδιαιτήματα/υποστρώματα στην Ελλάδα, συμπεριλαμβανομένων των πρώτων αναφορών. Από αυτές μία καταγραφή θεωρείται νέα σε παγκόσμιο επίπεδο και μία σε ευρωπαϊκό. Ταξινομικές μονάδες που πρωτοαναφέρονται σε αμιγή δασικά οικοσυστήματα Abies cephalonica αποτελούν και πρώτες καταγραφές σε παγκόσμιο επίπεδο για το οικοσύστημα αυτό.Με βάση τον Κατάλογο Συλλογών Καλλιεργειών, που εκδίδεται από το Παγκόσμιο Κέντρο Δεδομένων για τους Μικροοργανισμούς WDCM (2003), τα στελέχη 6 ειδών είναι μοναδικά παγκοσμίως, ενώ επιπλέον 4 στελέχη υπάρχουν κατατεθειμένα σε μία ή δύο άλλες Συλλογές Καλλιεργειών.Οι σημαντικότερες ταξινομικές παρατηρήσεις που έχουν προκύψει από τη μακροσκοπική και μικροσκοπική μελέτη των δειγμάτων μας είναι οι εξής: Υπάρχει σημαντική ποικιλομορφία και διακύμανση των μακροσκοπικών και μικροσκοπικών χαρακτήρων από άτομο σε άτομο ενός δείγματος και από δείγμα σε δείγμα μέσα σε ένα είδος, όπως π.χ. σε είδη του γένους Lentinellus και Rhodocybe.Ενίοτε καταγράφονται αποκλίσεις από τις βιβλιογραφικές περιγραφές, από τις οποίες οι σημαντικότερες αφορούν στα βασιδιοσπόρια (μέγεθος και μορφολογία), καθώς επίσης και στα κυστίδια και στην επιδερμίδα του πίλου, όπως π.χ. στα είδη Clitopilus prunulus, Rhodocybe gemina.Σε κάποια είδη καταγράφηκαν μικροσκοπικά γνωρίσματα, που δεν έχουν αναφερθεί προηγουμένως, όπως σε είδη των γενών Amanita και Limacella, στα οποία η εμφάνιση παχύτοιχων βασιδίων και βασιδιοσπορίων είναι χαρακτηριστική και πιθανολογείται ότι αποτελούν προσαρμογή στην επίδραση περιβαλλοντικών παραγόντων.Αρκετά από τα είδη ανήκουν σε ομάδες συγγενικών ειδών με παραπλήσιο ή εν μέρει επικαλυπτόμενο εύρος στις ιδιότητες των μορφολογικών χαρακτήρων, γεγονός που καθιστά ασαφή τα όρια διάκρισής τους και αναγκαία την περαιτέρω συγκριτική μελέτη με άλλες μεθόδους. Παράδειγμα αποτελούν τα τμήματα Vaginatae στο γένος Amanita, Dapetes στο γένος Lactarius και Omphalodei στο γένος Lentinellus.Το ενδιαίτημα ή το υπόστρωμα από το οποίο συλλέχθηκε ένα είδος μπορεί συνήθως να θεωρηθεί χαρακτηριστικό, αλλά δεν αποτελεί πάντα ασφαλές συμπληρωματικό κριτήριο για τον προσδιορισμό ή διαχωρισμό συγγενικών ειδών. Η διάκριση συγγενικών ειδών, όπως των Lactarius deliciosus και L. salmonicolor με βάση κυρίως την εμφάνισή τους σε δάση Pinus spp. και Abies spp. αντίστοιχα, θεωρείται επισφαλής. Είδη όπως τα Amanita phalloides, Lactarius chrysorrheus, Lactarius aurantiacus και Hohenbuehelia auriscalpium εμφανίζονται, αποκλειστικά το τελευταίο ή συνήθως τα πρώτα, σε φυλλοβόλα δάση, ενώ στην παρούσα μελέτη απαντούν σε αμιγή δάση Abies cephalonica.Επιπλέον, από τη μελέτη χαρακτηριστικών των απομονώσεων σε καθαρή καλλιέργεια συμπεραίνονται τα παρακάτω:Ο σχηματισμός αναμορφών και τελειομορφών σε ημισυνθετικό υπόστρωμα δεν είναι σύνηθες φαινόμενο και, όπου αυτά εμφανίζονται, θεωρούνται σημαντικά ταξινομικά γνωρίσματα. Αναμορφές, όπως κονίδια, χλαμυδοσπόρια και σκληρώτια, και τελειομορφές παρατηρήθηκαν σε 16 ταξινομικές μονάδες, στις 10 από τις οποίες καταγράφονται για πρώτη φορά διεθνώς.Τα μακροσκοπικά και μικροσκοπικά γνωρίσματα των αποικιών σε καλλιέργεια μπορούν να διαφέρουν από είδος σε είδος ενός γένους (π.χ. του Lentinellus) ή να εμφανίζουν σημαντικές ομοιότητες, που να χαρακτηρίζουν το γένος αυτό (π.χ. Hohenbuehelia, Pleurotus). Σε κάποιες περιπτώσεις, μπορούν να οδηγήσουν σε διαχωρισμό των ειδών σε ομάδες, οι οποίες συμπίπτουν με ταξινομικές μονάδες μέσα στο γένος (π.χ. Amanita). Μερικές φορές τα γνωρίσματα των αποικιών είναι τόσο χαρακτηριστικά και σταθερά, που μπορεί να θεωρηθεί ασφαλής η αντιστοίχιση και ο προσδιορισμός του είδους ή του γένους, από τα οποία έχουν προέλθει, χωρίς την παρουσία βασιδιοκαρπίων. Τέλος, τα γνωρίσματα αυτά συγκρινόμενα εμφανίζουν ενίοτε ομοιότητες σε επίπεδο γένους ή μεγαλύτερων ταξινομικών ομάδων υποδηλώνοντας πιθανές σχέσεις μεταξύ τους.Υπάρχουν μικροσκοπικά γνωρίσματα των βασιδιοκαρπίων, όπως η παρουσία κρίκων και διαφοροποιημένων υφών, που παραμένουν σταθερά και εμφανίζονται και στις απομονώσεις τους σε καλλιέργειες.Σε αρκετά στελέχη είναι εμφανής η παραγωγή εγχρώμων συνήθως μεταβολιτών, που είτε διαχέονται στο υπόστρωμα είτε εκκρίνονται στην επιφάνεια της αποικίας με μορφή σταγονιδίων. Συχνά η παραγωγή των μεταβολιτών ή η ποσότητά τους φαίνεται να συνδέεται με το σχηματισμό βασιδιοκαρπίων ή σκληρωτίων, καθώς και με καταστάσεις καταπόνησης (stress), όπως αντίξοες συνθήκες ανάπτυξης ή ανταγωνισμού.Αρκετά στελέχη βρέθηκαν να παράγουν λακκάση, γεγονός που αποτελεί ένδειξη της λιγνινολυτικής ικανότητάς τους. Σε λίγα σχετικά στελέχη ανιχνεύθηκε τυροσινάση. Η παραγωγή των ενζύμων αυτών φαίνεται να συσχετίζεται με συγκεκριμένες ταξινομικές μονάδες, ενώ η ποιοτική και ποσοτική δράση τους να εξαρτάται από το στέλεχος.Τα αποτελέσματα της παρούσας έρευνας εμπλουτίζουν τα δεδομένα που αφορούν στην ποικιλότητα των Βασιδιομυκήτων στην Ελλάδα και συμβάλλουν στην πληρέστερη γνώση των διαφόρων ταξινομικών μονάδων και στην επίλυση ταξινομικών προβλημάτων των Agaricales
