The role of human endogenous retroviruses in Graves disease

One of the most impressive early findings during the human genome mapping project was the discovery that 5-8% of the human genome was composed of human retroviruses (HERVs) and sequences derived from them. Frequently only partial sequences from HERVs (Long Terminal Repeats) have been identified that they could regulate the expression of adjacent genes. Two of these LTR5, LTR3 and LTR13 into consideration have been associated with immunological diseases. Taking the common origin of autoimmunity we tried to investigate the presence or absence of LTRs in patients with Graves’ disease. We observed that only LTR3 sequence was identified in Greek population and preliminary data suggests that LTR3 increases susceptibility for GD when inserted in either DRB1*-11 or DQB1*-0301 or DQA1*-0505 genotype. Our report is therefore in agree with other previous studies showing that human endogenous retrovirus particles can be considered as possible genetic markers.Κατά την τελευταία δεκαετία, διαπιστώθηκε ότι το ανθρώπινο γονιδίωμα περιέχει μεγάλο αριθμό αλληλουχιών από ανθρώπινους ενδογενείς ρετροϊούς ή με συσχετιζόμενες με αυτούς αλληλουχίες, ορισμένες από τις οποίες ενδεχομένως να έχουν και λειτουργική σημασία. Φαίνεται ότι οι HERVs (Human Endogenous Retroviruses) αντιπροσωπεύουν υπολείμματα προγενέστερης λοίμωξης από λοιμογόνο εξωγενή ρετροϊό, τα οποία ενσωματώθηκαν στην γενετική αλυσίδα. Μεμονωμένα τέτοια τμήματα ρετροϊών έχουν ενσωματωθεί σε τμήματα της HLA-DQB1* υποπεριοχής και έχουν μέχρι σήμερα συσχετισθεί με αυτοάνοσα νοσήματα. Σκοπός της εργασίας αυτής είναι η μελέτη για πρώτη φορά της συσχέτισης δύο τέτοιων τμημάτων LTR3 και LTR13 σε ασθενείς με νόσο Graves ελληνικής καταγωγής και εθνικότητας. Διαπιστώσαμε ότι η LTR3 αλληλουχία παρατηρείται σε αυξημένο ποσοστό ασθενών έναντι μαρτύρων και κυρίως σε ομόζυγη μορφή. Τα ευρήματα μας συμφωνούν με προηγούμενες μελέτες ότι οι ρετροϊοί μπορούν να χρησιμοποιηθούν ως ανεξάρτητοι γενετικοί δείκτες για ορισμένα αυτοάνοσα νοσήματα
