Biochemical and microbiological changes during a submerged chickpea fermentation (chickpea-starter) for the production of the traditional bread eftazimo

The microbiological and biochemical changes during a submerged fermentation of coarsely grounded chickpea seeds for 18 h, for the production of a chickpea-starter to be used as a leavening agent for the traditional bread “eftazimo”, as well as the heterogeneity of the strains involved in the fermentation were studied. For the biochemical study, changes in enzyme activities (cellulase, α-galactosidase, invertase, amylase and proteinase), changes in pH and acidity, liberation of FFA, FAA and reducing sugars, degradation of starch and protein (SDSPAGE), determination of mono- di- and oligo-saccharides (HPLC) of the fermented liquid during the submerged fermentation have been explored, as well as the safety of the fermented liquid. For the microbiological examination, the development of the microflora during the fermentation for 18 h (primary starter) and during raising a dough from wheat flour (adapted starter) in 2-hours intervals was first studied. Bacteria involved in the fermentation were isolated and characterized with phenotypic criteria (morphological and biochemical characters, SDS-PAGE of the whole cell protein). Selected strains were screened for carbohydrate fermentation ability and enzyme activities with traditional methods (amylase, invertase, proteinase, cellulase) as well as by the API ZYM system (19 different enzymes) and were typed by RAPDPCR. Cellulase, α-galactosidase, invertase, amylase and proteinase activities were detected in the fermented liquid and maxima were recorded at 10, 12, 8, 10 and 16 h of fermentation, respectively. A gradual decrease in pH (p<0.05) and an increase in acidity (p<0.05) were observed from 10 h onwards. Similarly, the concentration of free fatty acids increased (p<0.05), but appreciably after fermentation for 10 h. The amount of reducing sugars increased (p<0.05) on fermentation for 12 h and then gradually decreased (p<0.05). The α-galactosides also declined with fermentation and starch solubilization-degradation was detected after 8 h. The free amino acid content increased on fermentation, and protein degradation was evident after 8 h of incubation. In the fermenting liquid, only populations of Bacillus and Clostridium developed. Bacilli increased their loads significantly (p<0.05) during fermentation for 8-12 h and then remained constant. Clostridia developed (p<0.05) subsequently to levels of 10⁷ cfu/ml at 18 h, when the pH of the fermenting liquid had decreased (p<0.05) to ~ 5.4. It also seems that the rise of the adapted starter within 2 h was caused by enzymes present in the primary starter and those liberated after cell death by the declining populations of bacilli and clostridia. The principal groups of isolates in all fermentation experiments (with chickpea seeds from five different areas) seemed to have the phenotypic characteristics of B. cereus group and Cl. perfringens. SDS-PAGE of whole-cell proteins elucidated the taxonomic position of the B. cereus group of strains as B. cereus and B. thuringiensis and confirmed the phenotypic allocation of Cl. perfringens isolates. Strains phenotypically characterized as B. licheniformis and Cl. beijerinckii were also found to belong to these same species by SDS-PAGE. In addition, results showed that the fermenting broth was not toxic to mice when inoculated intraperitoneally and the product can thus be considered as safe for consumption. The results showed that bacilli exhibited amylase, invertase and proteinase activities. With the API ZYM system Cl. perfringens strains showed strong α-glucosidase, α-galactosidase, alkaline phosphatase, phosphohydrolase and esterase activities, medium esterase/lipase, β-galactosidase activities; some strains exhibited strong β-glucuronidase activities and weak or no leucine-, valine- and cysteine-arylamidase activities. B. cereus strains showed strong acid phosphatase activities, medium alkaline phosphatase, esterase, esterase/lipase, leucine arylamidase, phosphohydrolase and α-glucosidase activities and weak valine-, trypsin and α-chymotrypsin activities. API-ZYM proved to be a practical system for the rapid differentiation of Cl. perfringens from Cl. beijerinckii. Furthermore, RAPD-PCR results revealed a high heterogeneity in B. cereus strains and suggest the usefulness of RAPD-PCR for the differentiation of Cl. perfringens and Cl. beijerinckii isolates. Overall, the indigenous microbial population of the chickpea fermentation liquid may cause changes in its chemical constituents that can be attributed mainly to bacilli, until 8-10 h of fermentation, and to clostridia thereafter.Μελετήθηκαν οι μικροβιολογικές και βιοχημικές μεταβολές κατά τη διάρκεια της ζύμωσης βυθού χονδροαλασμένων σπόρων ρεβιθιού (ρεβιθομαγιά) για την παρασκευή του παραδοσιακού εφτάζυμου ψωμιού. Οι βιοχημικές μεταβολές που μελετήθηκαν κατά την πορεία της ζύμωσης των σπόρων ρεβιθιού για 18 ώρες, ήταν η παρακολούθηση της δραστικότητας ενζύμων της υγρής φάσης (αμυλάση, πρωτεϊνάσες, κυταρινάση, α-γαλακτοσιδάση, ιμβερτάση) η μεταβολή του pH, της οξύτητας, των λιπαρών οξέων, η ελευθέρωση αμινοξέων και αναγόντων σακχάρων, η αποικοδόμηση του αμύλου και των πρωτεϊνών (SDSPAGE), ο προσδιορισμός των μονο-, δι- και ολιγοσακχαριτών με υγρή χρωματογραφία υψηλής απόδοσης (HPLC). Επίσης ελέχθηκε η ασφάλεια του υγρού της ζύμωσης με ενδοπεριτοναϊκή έγχυση σε ποντίκια. Για τη μελέτη των μικροβιολογικών μεταβολών μελετήθηκε η ανάπτυξη της μικροχλωρίδας, κατά τη διάρκεια της ζύμωσης των σπόρων για 18 ώρες, καθώς και κατόπιν ανάμιξής του ζυμωμένου υγρού με αλεύρι σίτου για την παραγωγή μιας προσαρμοσμένης καλλιέργειας, η οποία επωάστηκε για 2 ώρες. Απομονώθηκαν οι υπεύθυνοι για τη ζύμωση μικροοργανισμοί, ταξινομήθηκαν με φαινοτυπικά κριτήρια (μορφολογικοί και βιοχημικοί χαρακτήρες, ηλεκτροφόρηση πρωτεϊνών ολόκληρου του κυττάρου με SDS-PAGE). Στη συνέχεια μελετήθηκε η ετερογένεια επιλεγμένων στελεχών με γενοτυπικά κριτήρια (RAPD-PCR), την ικανότητα ζύμωσης σακχάρων, την παραγωγή ενζύμων (αμυλάση, κυτταρινάση, πρωτεϊνάσες, α-γαλακτοσιδάση) με εργαστηριακές μεθόδους, καθώς και 19 ακόμη ενζύμων με το σύστημα ΑPI-ZYM. Η πτώση του pH και η αύξηση της οξύτητας ήταν σημαντική (p<0,05) από τη 10η ώρα της ζύμωσης και μετά. Επίσης τα ελεύθερα λιπαρά οξέα αυξήθηκαν σημαντικά (p<0,05) από τη 10η ώρα και έπειτα. Τα ανάγοντα σάκχαρα αυξάνονταν έως τη 12η ώρα και μετά σταδιακά μειώνονταν. Τα ελεύθερα αμινοξέα αυξάνονταν συνεχώς έως το τέλος της ζύμωσης, οι α-γαλακτοζίτες μειώθηκαν με την πορεία της ζύμωσης και η αποικοδόμηση των πρωτεϊνών ήταν εμφανής μετά τη 8η ώρα της ζύμωσης. Η κυτταρινάση ήταν στο μέγιστο της δραστικότητάς της την 10η ώρα, η α-γαλακτοσιδάση την 12η, η ιμβερτάση την 8η και η αμυλάση την 10η ώρα. Οι πρωτεϊνάσες ανιχνεύτηκαν από την 12η ώρα και εμφάνισαν ένα μέγιστο την 16η ώρα. Στο υγρό της ζύμωσης αναπτύχθηκαν μόνο πληθυσμοί σποριογόνων βακτηρίων, βάκιλοι και κλωστρίδια. Οι βάκιλοι αυξάνονταν σημαντικά (p<0,05) κατά τις πρώτες 8-10 ώρες της ζύμωσης φτάνοντας σε επίπεδα 10⁶ cfu/ml, και στη συνέχεια έως το τέλος της ζύμωσης αναπτύσσονταν με μικρότερο ρυθμό, φτάνοντας σε τελικά επίπεδα 10⁷ - 10⁸ cfu/ml. Τα κλωστρίδια αναπτύσσονταν με αργό ρυθμό έως τις 10 ώρες (10³ cfu/ml) και με γρηγορότερο ρυθμό στη συνέχεια, ενώ το pH στο υγρό της ζύμωσης μειώθηκε στο 5,4 (p
