Endothelial cells (EC), the cells that line blood vessels, can be activated by proangiogenic cues and rearrange their cell-cell junctions to allow for sprouting angiogenesis. Bone morphogenetic protein (BMP) signaling plays a fundamental role in regulating angiogenesis and is pro-angiogenic through ligands BMP2 and BMP6. The BMP pathway is negatively regulated by SMAD6, an intracellular inhibitory SMAD. SMAD6 is anti-angiogenic and modulates EC adherens junction turnover both in vivo and in vitro.
