AbstractObjective Quantitation and analyses of the fine structure of glycosaminoglycans are increasingly important for understanding many biological processes, including those most critical for understanding skeletal biology. We have developed a novel procedure, fluorophore-assisted carbohydrate electrophoresis (FACE), for determination of glycosaminoglycan fine structure and estimation of chain length.Design FACE utilizes enzymes that cleave glycosaminoglycans to create products, usually disaccharides, characteristic of the enzyme specificity. Each cleavage exposes a new reducing terminus that is fluorotagged by reductive amination with 2-aminoacridone.
