AbstractHuman erythrocyte spectrin was labelled with the probe 5,5′-disulfato-1-(6-hexanoic acid N-hydroxysuccinimide ester)-1′-ethyl-3,3,3′,3′-tetramethylindocarbocyanine (Cy3). Cy3-spectrin was bound to the outer surface of dimyristoylphosphatidylcholine (DMPC) multilamellar vesicles and its diffusion measured by fluorescence recovery after photobleaching (FRAP). It was found that at 30°C, above the lipid gel to liquid-crystalline phase transition of the lipids, Cy3-spectrin had an unexpectedly high diffusion coefficient D=(2.1±0.6)×10−7 cm2/s.
