ELISA was used to determine the epitopes of several HIV-1 NAbs by testing binding to SF162 wt Env, SF162 ΔV1, SF162 ΔV2, SF162 ΔV3, and SF162 D368R mutants. (A) Binding phenotype of V3-specific mAbs determined by ELISA. (B) Binding phenotype of CD4-specific mAbs determined by ELISA. Using a comprehensive library of HIV-1 Env mutants, cytometric flow sorting was used to identify critical residues for the binding of HIV-1 mAbs.
