Inhaled delivery of polymyxins is important for respiratory infections caused by Gram-negative bacteria that are resistant to almost all current antibiotics. Unfortunately, the current dosage regimens have not been optimised yet using PK/PD, which may result in potential respiratory toxicity. The goal of this thesis is to investigate the mechanisms of polymyxin-induced respiratory toxicity and develop safer therapies using cell biology and systems biology approaches
