Structure and Dynamics of Photosynthetic Membranes as Revealed by Neutron Scattering

Un des plus importants challenges scientifique de notre époque, est de trouver des réponses et solutions pour les questions et problèmes différents concernant la consommation d'énergie d'humanité. Les études scientifiques sur l'utilisation des ressources renouvelables spécialement d'énergie solaire gagnent en importance. Dans la biosphère conversion d'énergie solaire est accompli par des organismes photosynthétiques. Photosynthèse, spécialement ses mécanismes différents de régulation sont loin d'être compris. Les membranes thylacoïdales possèdent un rôle central dans la photosynthèse. Dans cette thèse nous étudions les paramètres structuraux des membranes thylacoïdales isolé des plants ou dans les algues avec la diffusion de neutrons aux petits angles et la dynamique des fragments des membranes thylacoïdales (BBY) avec diffusion élastique incohérente des neutrons. Des résultats présentés ici, présentent évidence pour l'occurrence des réorganisations petites mais bien discernables des membranes thylacoïdales pendent la photosynthèse et pour une transition dans les membranes BBY à les températures physiologiques et aussi présentent des exemples pour l'utilisation possible de diffusion neutronique pour l'investigation des échantillons biologiques in vivo.One of the most important scientific challenges of our time is to find answers and solutions for the various questions and problems about the energy consumption of humanity. Scientific studies on the utilization of renewable resources, especially of solar energy, gain more and more importance. In the Biosphere, solar energy conversion to chemical energy is performed by photosynthetic organisms. Photosynthesis, especially its different regulatory mechanisms is far from being understood. Thylakoid membranes possess a central role in photosynthesis. In the present thesis we investigate structural parameters of the thylakoid membranes, isolated from higher plants and in different algal cells, with small-angle neutron scattering, and the dynamics of isolated thylakoid membrane fragments with elastic incoherent neutron scattering. The presented results, in addition to providing clear evidence for the occurrence of small but well discernible membrane reorganizations in thylakoid membranes during photosynthesis, and for a transition in BBY membranes at physiological temperatures, also present example of possible application of neutron scattering for the investigation of in vivo biological samples
