The purpose of this experiment was to observe ICP22's effects on expression of immediate early, early, and late genes of herpes simplex virus-1. This was tested through GFP fluorescence of reporter plasmids after infection with the KOS wild type or the n199 ICP22 null virus. ICP22 suppressed the activity of six of the eight promoters tested, which were ICPO, ICP22/47, ICP27, TK, gC, and VP5. The two viral promoters that did not show suppression were ICP4 and ICP6. We conclude that ICP22 is not a universal repressor, but rather specifically targets certain viral genes and directly regulates them
