Vektoren des Adeno-Assoziierten Virus (AAV) zeichnen sich durch eine Reihe von vorteilhaften Eigenschaften aus, wie beispielsweise einer geringen Immunanwort, ein breites Wirtszellspektrum sowie die Fähigkeit sowohl ruhende als auch sich teilende Zellen infizieren zu können. Aufgrund dieser Eigenschaften sind rekombinante AAV-Vektoren (rAAV) geradezu als Gentherapievektoren prädestiniert. Ziel meiner Arbeit war die Etablierung eines rAAV-vermittelten Gentargeting-Systems, das als proof of principle System verwendet wurde, um ein defektes egfp-Gen über Homologe Rekombination (HR) direkt am mutierten Ziellokus korrigieren zu können.
