The extracellular matrix of fibrotic liver tissue stores the proforms of gelatinases by binding to the tripelhelical structure of collagens

Bei der Induktion, Aufrechterhaltung und Auflösung der Leberfibrose spielen neben direkten zellulären Interaktionen die Wechselwirkungen zwischen den verschiedenen Leberzellen und der extrazellulären Matrix (EZM) eine zentrale Rolle. Dabei wirken EZM-Bestandteile, wie z.B. die Kollagene, nicht nur direkt auf die Zellen, sondern modulieren deren Aktivität auch indirekt durch die Speicherung von anderen Molekülen, wie z.B. Wachstumsfaktoren (Crinopexy). Neben Wachstumsfaktoren wurde auch für verschiedene Matrixmetalloproteinasen (MMPs), welche wesentlich am physiologischen und pathologischen Umbau der EZM beteiligt und therapeutisch von großem Interesse sind, eine Speicherung in der EZM hypothetisiert. Basierend darauf war Zielsetzung dieser Arbeit, die Interaktion bzw. die Speicherung von MMPs in der EZM weiter zu untersuchen, potentielle Bindungsstellen auf den Hauptkomponenten der EZM, nämlich den Kollagenen, zu analysieren und diese Bindung ggf. direkt - z.B. mit Peptidanaloga - zu modulieren. Immunhistologisch wurde in situ (APAAP) in fibrotischem Lebergewebe nachgewiesen, dass endogene MMPs auch außerhalb der produzierenden Zellen im Gewebe gebunden vorliegen. In Festphasenassays (96-Lochplatten, dot-blot) banden [125I]-markierte proMMPs bevorzugt an die Kollagene Typ I und III aber nur schwach an andere untersuchte EZM- Bestandteile. Die Proformen beider Enzyme wurden im Gegensatz zu den aktivierten Formen sehr stark durch die gereinigten Matrixmoleküle gebunden. Damit wurde erstmals gezeigt, dass die Bindung von Gelatinasen an Nichtsubstrat-Kollagene vom Aktivierungszustand des MMPs abhängig ist, die EZM also einen Speicher für die latenten Gelatinasen darstellt. Des Weiteren konnte in einem fluorimetrischen Aktivitätsassay für MMP-9 gezeigt werden, dass die Aktivierung selbst ebenfalls durch die Bindung an Kollagen unterdrückt wird. Die Bindungsversuche mit definierten Fragmenten von KI sowie synthetischen Kollagenmimetika, die sich im Wesentlichen durch den Anteil und die Rigidität vorhandener tripelhelikaler Strukturen unterschieden, zeigten, dass mit Zunahme des Anteils tripelhelikaler Strukturen (Zunahme in der Reihenfolge KI, α1-Kette von KI [α1(I)], CB8- bzw. CB7-Fragment von α1(I)), die Fähigkeit latente Gelatinasen zu binden, zunahm. So lag die Annahme nahe, dass nicht eine charakteristische Primärstruktur, sondern die Sekundärstruktur „kollagene Tripelhelix“ selbst als Konsensusstruktur für die Bindung der proMMPs fungiert. Dies wurde dadurch bestätigt, dass die Proformen beider Enzyme auch an das synthetische Kollagenpeptid Glycin-Prolin-Hydroxprolin (GPO)10 binden und (GPO)10 allein effizient die Bindung der proMMPs an immobilisiertes KI blockieren kann. Es konnte somit gezeigt werden, dass die tripelhelikale Struktur von Kollagenen die Konsensusbindungsstruktur für latente Gelatinasen darstellt. Durch Modulation mittels spezifischer (synthetischer) Peptide könnte es gelingen, diese latenten MMPs lokal zu aktivieren und somit die Fibrose/Zirrhose lokal im Sinne einer fibrolytischen Therapie zu modulieren.Interactions between different liver cells and the extracellular matric (ECM) play a central role in the initiation, perpetuation and resolution of liver fibrosis. Parts of the ECM not only interact directly with different liver cells but also modulate their activity by storage of other molecules like e.g. growth factors (crinopexy). Based on earlier findings it was hypothesized that the ECM functions as a reasonable storage site for matrix metalloproteinases (MMPs) which are essential for the remodelling of the extracellular matrix. Objective was to examine the interaction/storage of MMPs to the ECM and to identify possible binding sites and subsequently modulate the binding with collagen mimetics. Immunohistological staining (APAAP) of proMMP-2/-9 binding to the ECM emphasized a depot function of this non-cellular compartment in fibrotic tissues. Solid phase binding studies using clearly defined immobilized native collagens, type I collagen (CI) single chains, CI-derived peptides and synthetic peptides mimicking collagenous triple helical structures showed that the proMMPs and not the active gelatinases were directly retained by these ECM molecules. Binding strength was found to be enforced within the sequence: heterotrimeric CI, homotrimeric CI α-chains and a structural analog consisting of ten Gly-Pro-Hyp repeats ((GPO)10), thus confirming that not a primary structure but the secondary tripelhelical structure serves as a consensus motif for the proMMP-2/-9 binding to non- substrate collagens. In a fluorometric assay for MMP-9 it was shown that the activation itself can be suppressed by the binding to collagen.
