Establishment of the functional characterization of primary keratinocytes from adult epidermis for the examination of the nuclear transcription factor PPARδ in the pathogenesis of psoriasis vulgaris

Der Peroxisome proliferator activator receptor delta (PPARδ) ist ein nukleärer Rezeptor aus der Superfamilie der Steroidhormonrezeptoren. Die molekularen Funktionen von PPARδ sind komplex und beinhalten sowohl die β-Oxidation von Fettsäuren, die Differenzierung humaner epithelialer Stammzellen als auch die Proliferation von humanen Keratinozyten. Hinsichtlich einer Schlüsselfunktion von PPARδ in der Differenzierung humaner Keratinozyten und einer möglichen Beteiligung des Rezeptors an der Pathogenese der Psoriasis vulgaris finden sich in der Fachliteratur seit Ende der 1990er Jahre gesicherte Ergebnisse. Zur genaueren experimentellen Untersuchung der Funktion von PPAR in der Psoriasispathogenese wurde im Rahmen dieser Promotionsarbeit eine Co- Zellkultur aus humanen Fibroblasten als Feeder-Layer und humanen Keratinozyten aus psoriatischer Haut etabliert. Dabei wurde das allgemein gängige Kulturverfahren nach Rheinwald und Green durch den bewussten Verzicht auf murine Fibroblasten als Feeder-Layer-Zellen modifiziert und optimiert. Diese Zellkultur wurde nachfolgend verwendet, um die funktionelle Rolle von PPARδ in primären Keratinozyten zu untersuchen. Hierzu wurden primäre Keratinozyten mit einem spezifischen Liganden für PPARδ aktiviert und daraufhin die globale Genexpression mittels Mikroarray Analysen bestimmt. Dabei konnte die metabolische Funktion von PPARδ in Keratinozyten anhand der aktivierten Zielgene nachgewiesen werden. Weiterhin wurde der Transkriptionsfaktor Heparin binding epidermal growth factor (HB-EGF) als Zielgen von PPARδ identifiziert. Somit konnte PPARδ als Mitverursacher der langjährig bekannten Überexpression von HB-EGF in psoriatischer Epidermis und der durch diese induzierten keratinozytären Hyperproliferation charakterisiert werden. Die im Rahmen dieser Arbeit erzielten Ergebnisse deuten auf eine wichtige Rolle von PPARδ als Schlüsselregulator bei der Übersetzung des vor allem durch T-Lymphozyten erzeugten entzündlichen Mikromilieus innerhalb der Epidermis in den typischen kutanen psoriatischen Phänotyp hin. Ferner wurde eine Methode zur Transduktion von humanen Keratinozyten aus psoriatischer Haut mit lentiviralen shRNA- tragenden Vektoren etabliert. Mittels des hier vorgestellten Protokolls zur lentiviralen Transfektion können bei primären psoriatischen Keratinozyten Transduktionsraten von über 70% erreicht werden, wobei die zur Transfektion eingesetzten Virussuspensionen Titer bis zu 106 Viren pro ml aufwiesen. Die erfolgreiche Einbringung und Expression der shRNA in die Keratinozyten konnte auch funktionell mittels RT-PCR nachgewiesen werden. Die Etablierung einer Methode zur erfolgreichen Transduktion primärer humaner Keratinozyten mittels niedrigtitrigen Virususpensionen erlaubt eine effektive Anwendung dieser Methode ohne Aufkonzentration der Virusüberstände.The Peroxisome proliferator activator receptor Delta (PPARδ) is a nuclear receptor protein belonging to the family of the steroid hormone receptors. Its molecular functions are of great complexity and include the stimulation of the β-oxidation of fatty acids, the terminal differentiation of epithelial stem cells as well as the proliferation of human keratinocytes. Scientific evidence regarding PPARδ as a key player in the cellular differentiation of human keratinocytes and indications of a possible link to the complex pathogenesis of psoriasis vulgaris is found in the scientific literature as of the late 1990s. For a more exact characterization of the physiological functions of PPARδ in the pathogenesis of psoriasis vulgaris, a co-cell culture of human fibroblasts as feeder layer and primary human keratinocytes from psoriatic skin was established. As a result, the common culture technique of Rheinwald and Green was modified and optimized by the deliberate relinquishment of murine fibroblasts as feeder cells. This new cell culture was further used to study the functional diversity of PPARδ in primary human keratinoytes. Therefore the global gene expression profile in primary keratinocytes was studied by microarray after activation of the cells with a specific PPARδ ligand. This approach allowed detection of specific target genes of PPARδ. In particular, the transcriptional factor Heparin binding epidermal growth factor (HB-EGF) could be identified as a target gene of PPARδ. Due to these findings PPARδ can be regarded as an important causal agent of the well-known overexpression of HB-EGF in the psoriatic epidermis and the resulting hyperproliferation of psoriatic keratinocytes. The gained results point toward a pivotal role of PPARδ as a regulative agent in the transformation of a primarily T cell-derived, proinflammatory microenvironment into the characteristic psoriatic phenotype. Furthermore, a new method for the transduction of human keratinocytes with lentiviral vectors carrying shRNAi was established. With the presented protocol transfection, rates of up to 70% could be achieved, while the applied supernatants showed relatively low viral titers of up to 106 viral particles per millilitre. The successful intracellular insertion and genetic expression of the shRNAi elements could be shown by RT-PCR.
