Active zones are highly-specialized sites in the presynaptic bouton that are essential for neurotransmitters release. The molecular machinery mediating the fusion of synaptic vesicles (SVs) at presynaptic active zone membranes has been studied in detail, and several essential components have been identified. Active zone associated protein scaffolds were so far viewed as rather modulatory for transmission.
