Association between polymorphisms of the GABAB\- and the Dopamine-D2 receptor genes and alcohol dependence

Im Rahmen der vorliegenden Arbeit wurden drei Polymorphismen des DRD2-Rezeptors sowie drei Polymorphismen des GABA-B-R1-Rezeptors jeweils im Patienten- und Kontrollkollektiv genotypisiert und ihr Einfluss auf die Alkoholabhängigkeit untersucht. Der Exon8-Polymorphismus zeigte sowohl bezüglich der Genotyp-Häufigkeiten als auch bezüglich der Allel-Häufigkeiten signifikante Unterschiede zwischen dem Patienten- und dem Kontrollkollektiv (p<0,05). Dagegen konnten bei der Untersuchung des -141C und des TaqI-A-Polymorphismus weder beim gesamten Patientenkollektiv noch bei den untersuchten Subgruppen signifikante Assoziationen entdeckt werden. Der Genotyp [InsIns/GG/A2A2] zeigte eine signifikant erhöhte Häufigkeit in der Patientengruppe mit einer Odds Ratio von 4,59 (p<0,01). Die Untersuchung der geschätzten Haplotypen zeigte für den Haplotypen [Ins/G/A2] ein signifikant höheres Vorkommen in der Patientengruppe, wobei die Odds Ratio jedoch nur bei 1,54 liegt (p<0,01). Dagegen war der Haplotyp [Del/A/A2] in der Patientengruppe signifikant seltener vorhanden, bei einer Odds Ratio von 0,396 (p<0,05). Die Polymorphismen -141C-Ins/Del und Exon8-A/G zeigten sowohl in der Patienten- als auch in der Kontrollgruppe ein Kopplungsungleichgewicht. Dagegen bestand für die Polymorphismen -141C und TaqI-A sowie die Polymorphismen Exon8 und TaqI-A ein Kopplungsungleichgewicht nur in der Patientengruppe, nicht aber in der Kontrollgruppe. Die vorliegende Studie erbrachte keinen Hinweis, dass einer der drei untersuchten GABA-B-R1-Rezeptor-Polymorphismen einen substantiellen Einfluss (Odds Ratio>3) auf die Pathogenese der Alkoholabhängigkeit ausübt. Dies gilt sowohl für die Gesamtheit der untersuchten Alkoholiker als auch für die untersuchten Subgruppen. Allerdings fanden sich beim Exon7- und Exon11-Polymorphismus signifikante Assoziationen zwischen den Allel-Häufigkeiten und der Gruppe der Alkoholiker mit antisozialer Persönlichkeitsstörung. Obwohl die vorliegende Studie bei den untersuchten Polymorphismen einige signifikante Assoziationen zur Alkohol- Abhängigkeit bzw. zu der untersuchten Subgruppe der Patienten mit dissozialer Persönlichkeitsstörung aufweist, können aufgrund der Vielzahl der untersuchten Zusammenhänge und der daraus resultierenden Gefahr falsch-positiver Ergebnisse keine eindeutigen Zusammenhänge abgeleitet werden.Genotypic variants of the DRD2-receptors and the GABA-receptor possibly possess an influence on the prevalence and the development of alcohol dependence. In this study three polymorphisms of the DRD2-receptors and three polymorphisms of the GABA-B-R1-receptor were genotyped in a patient and a control population and its influence on alcohol dependence was examined. The Exon8-polymorphism showed in the genotype frequencies and in the allele frequencies significant differences between the patient and the control population (p<0,05). On the other hand significant associations couldn't be revealed in the investigation of the -141C- and the TaqI-A-polymorphism neither at the entire patient population nor with the examined subgroups. The genotype [InsIns/GG/A2A2] was significantly more frequent in the group of patients with an Odds Ratio of 4,59 (p<0,01). The investigation of the estimated haplotypes showed a significantly higher occurrence of the haplotypes [Ins/G/A2] in the group of patients, whereas the Odds Ratio lies however only at 1,54 (p<0,01). On the other hand the haplotype [Del/A/A2] in the group of patients, with an Odds Ratio of 0,396 (p<0,05) was significantly less frequent. The polymorphisms -141C-Ins/Del and Exon8-A/G showed both in the patient and in the control's group a linkage disequilibrium. Regarding the polymorphisms -141C and TaqI A as well as the polymorphismen Exon8 and TaqI-A a linkage disequilibrium only in the group of patients could be observed. The results of this study did not show that one of the three examined GABA-B-R1 -receptor-polymorphisms exerts a substantial influence (Odds Ratio>3) on the pathogenesis of alcohol dependence. This applies both to the examined alcoholics and to the examined subgroups. However significant associations were present between the allele frequencies of the Exon7 and the Exon11-polymorphism and the group of the alcoholics with antisocial personality disorder. Although the available study shows some significant associations between the examined polymorphisms and alcohol dependence and to the examined subgroups of the patients with antisocial personality disorder, no clear relationship can be defined
