Analysis of the PI3K / AKT / mTOR pathway in squamous cell carcinoma (SCC)

Lungenkarzinome (LC) sind die am häufigsten zum Tode führenden Tumoren in Deutschland sowohl bei Männern als auch bei Frauen. Die Lungenkarzinome unterteilen sich in zwei histologische Hauptformen, die kleinzelligen (SCLC) und nicht kleinzelligen Lungenkarzinome (NSCLC). Nicht kleinzellige Lungenkarzinome machen den Großteil der LC aus und lassen sich histologisch weiter in Adenokarzinome (AC), Plattenepithelkarzinome (SCC) und großzellige Karzinome (LCC) subtypisieren. Plattenepithelkarzinom machen ca. 30% alle Nicht kleinzelligen Lungenkarzinome aus. Molekulare Veränderungen in Plattenepithelkarzinome sind für mehr als 50% der Fälle beschrieben. Viele Dysregulationen und Mutationen in PI3K/AKT/mTOR-Kaskade bei Plattenepithelkarzinom sind bereits identifiziert. Der PI3-Kinase/AKT/mTOR- Signalweg ist bei vielen Tumoren direkt oder indirekt in die Malignomgenese involviert. Verschiedene PI3K/AKT/mTOR-Signalweg Inhibitoren wie Rad001, NVP- BEZ235 und NVP-BKM120 sind bereits als alleinige Target-Therapie oder in Kombination mit der Chemotherapie in vielen Karzinome in klinischer Entwicklung. In dieser Arbeit wurde der Expressions- und Phosphorylierungsstatus der verschiedenen Proteine des PI3K/AKT/mTOR- Signalwegs mittels Western blot untersucht. Die Proliferationsmessung wurde durch MTT-Test, der Zelltod- und Zellarrest durch FACS analysiert. Die aktive Herrunterregulation von proapoptotischem Protein PUMA wurde durch Einbringen von PUMA-siRNA in den Zellen mittels eine Elektroporation durchgeführt. 6 Die Schlüsselproteine des mTOR-Signalwegs waren bei allen untersuchten SCC Zelllinien aktiv. Die untersuchten SCC Zellen konnten anhand der Proliferations- und Zelltodmessungen beim Einsatz von mTOR-Signalweg Inhibitoren in Rad001 resistente und sensitive SCC Zellen unterteilt werden. Der Dual-Inhibitor NVP-BEZ235 induziert bei allen SCC Zellen und insbesondere bei Rad001 sensitiven Zellen eine Proliferationshemmung und nur bei Rad001 sensitiven Zellen zusätzlichen Zelltod. Die weitere Untersuchung der Rad001 sensitiven SCC Zellen mittels Western Blot Analyse zeigte Heraufregulation des proapoptotischen Faktors PUMA und verstärkte CaspasenSpaltung. Eine Transfektion der zelltodempfindliche SCC Zellen mit PUMA-siRNA nach anschließender Kultivierung mit NVP-BEZ235 führte zum verringerten Zelltod und zur verminderten Expression von PUMA sowie im Western Blot eine verminderte Menge an gespaltener Caspase 3. Dies signifikante Reduktion der durchflusszytometrischen Zelltodmengen und parallel hierzu die Abnahme der Caspase 3 Spaltung zeigten die Bedeutung von PUMA für die Zelltodinduktion durch Hemmung des PI3K/AKT/mTORSignalwegs bei Plattenepithelkarzinom. Die vorliegenden Ergebnisse bezüglich der molekularen und funktionellen Bedeutung des PI3K/AKT/mTOR Signalweges deuten darauf hin, dass sich diese Signalwege als therapeutische Ziele in der Zukunft sehr gut könnten.Lung cancer (LC) ist the most frequent cause of cancer-related death in Germany in men and women alike. The lung cancer is divided into two main histological types, small cell (SCLC) and non-small cell lung cancer (NSCLC). Non small cell lung cancer make up the bulk of the LC and can be subtyped histologically further into adenocancer (AC), squamous cell cancer (SCC) and large cell cancer (LCC). Squamous cell lung cancer account for approximately 30% of all non-small cell lung cancers. Molecular alterations in squamous cell cancer have been described for more than 50% of cases. Various dysregulation and mutations in PI3K/AKT/mTOR cascade in squamous have already been identified. The PI3K/AKT/mTOR pathway is directly or indirectly involved in the malignomgenesis in many tumors. Various PI3K/ AKT/mTOR pathway inhibitors like Rad001, NVP-BEZ235 and NVP-BKM120 are already the single target agent or in combination with chemotherapy in many cancers in clinical development. In this study the expression and phosphorylation status of the various proteins of the PI3K/AKT/mTOR signaling pathway was investigated by western blot. The proliferation measurement was analyzed by MTT test, the cell death and cell arrest by FACS. The active sub-regulation of proapoptotic protein PUMA was carried out by introducing PUMAsiRNA into the cells by electroporation. 8 The key proteins of the mTOR pathway were active in all examined cell lines (SCC). The investigated SCC cells could be divided based on the proliferation and cell death when using mTOR signaling pathway inhibitors in Rad001 resistant and sensitive cells. The dual inhibitor NVP-BEZ235 induced in all SCC cells and particularly Rad001 sensitive cells proliferation inhibition and only in Rad001 sensitive cells additional cell death. The further study of Rad001 sensitive SCC cells by western blot analysis showed upregulation of the proapoptotic factor PUMA and enhanced caspase cleavage. Transfection of sensitive SCC cells with PUMA siRNA after subsequent cultivation with NVP- BEZ235 led to decreased cell death and in western blot decreased expression of PUMA and a reduced amount of cleaved caspase 3. This significant reduction of the in the cell death in FACS and parallel to this, the decrease in caspase 3 cleavage showed the importance of PUMA for cell death induction by inhibition of the PI3K / AKT / mTOR signaling pathway in squamous cell carcinoma. The present results concerning the molecular and functional significance of the PI3K/AKT/mTOR pathway suggests that these pathways could very well as therapeutic targets in the future
