Expression patterns of polo-like kinase 1 and 3 in human gastric cancer

Die Polo-like-Kinasen sind als zentrale mitotische Regulatoren am fehlerfreien Ablauf des Zellzyklus' beteiligt, indem sie u. a. die Zentrosomenorganisation, die Bildung des mitotischen Spindelapparates und die Zytokinese kontrollieren (PLK1), aber auch durch Mitwirkung an den Kontrollpunkten des Zellzyklus' antiproliferative Funktionen übernehmen (PLK2 und 3). In verschiedenen Tumorgeweben konnte eine Überexpression der PLK1 festgestellt werden, wohingegen zur Expression der PLK3 in Tumoren variierende Daten vorliegen. Weiterhin ließ sich sowohl in vitro als auch in vivo zeigen, dass eine Inhibition der PLK1 eine Induktion von Apoptose und eine Suppression des Tumorwachstums bewirkt. In der vorliegenden Arbeit wurden die Expressionsmuster der PLK1 und 3 im paraffineingebetteten Tumorgewebe von 135 Patienten mit Magenadenokarzinom sowie in 46 korrespondierenden Lymphknotenmetastasen immunhistochemisch untersucht. Die Expressionsdaten wurden mit klinisch-pathologischen Parametern und mit dem Patientenüberleben korreliert. Darüber hinaus erfolgte die Analyse der Expression von PLK1 in einer humanen Magenkarzinomzelllinie sowie kryokonserviertem Magentumorgewebe mit Hilfe von Immunoblot- und Immunfluoreszenztechniken. Eine Überexpression der PLK1 fand sich, verglichen mit normaler Magenschleimhaut, in 54,1% der Karzinome. Diese Überexpression korrelierte positiv mit dem Tumorstadium, dem Lymphknotenstatus und dem Alter der Patienten bei Diagnosestellung sowie einem diffusen Wachstumsmuster nach Laurén. Die PLK1-Expression im Primärtumor unterschied sich hierbei nicht wesentlich von der PLK1-Expression in den korrespondierenden Lymphknotenmetastasen. 57,3% der Karzinome zeigten eine PLK3-Überexpression, wobei das Expressionsniveau der PLK3 in den entsprechenden Lymphknotenmetastasen etwas höher lag. Die PLK3-Expression korrelierte positiv mit dem Tumorstadium und dem Lymphknotenstatus sowie mit der Expression der PLK1. Die univariate Überlebensanalyse zeigte eine signifikante negative prognostische Relevanz verstärkter PLK1- und PLK3-Expression, wohingegen die Expression beider Isoenzyme in der multivariaten Überlebensanalyse keinen unabhängigen prognostischen Einfluss hatte. Im Vergleich zu normaler Magenmukosa wiesen sowohl die untersuchte Magenkarzinomzelllinie als auch die kryokonservierten Magentumorproben eine gesteigerte PLK1-Expression auf. Zusammenfassend ließ sich in der vorliegenden Arbeit eine Korrelation der Expression von PLK1 und 3 im Magenkarzinom mit prognostisch ungünstigen klinisch-pathologischen Parametern sowie dem Patientenüberleben feststellen, was für eine Beteiligung dieser Proteine an der Entstehung des Magenkarzinoms spricht und insbesondere das PLK1-Molekül als Ziel neuer chemotherapeutischer Ansätze attraktiv erscheinen lässt.Polo-like kinases are important mitotic regulators controlling cell cycle progression by contributing to centrosome organisation, mitotic spindle formation and cytokinesis (PLK1). However, some members of this enzyme family also participate in cell cycle checkpoint functions (PLK2 and PLK3). Numerous tumours were shown to overexpress PLK1 whereas PLK3 expression varied in different tumour entities. Furthermore, inhibition of PLK1 in vitro and in vivo leads to apoptosis and suppression of tumour growth. In the present study, expression patterns of PLK1 and 3 were investigated in paraffin- embedded tumour tissue from 135 patients with gastric adenocarcinoma and in corresponding lymph node metastases using immunohistochemistry. Expression data were correlated with clinicopathological parameters and with patient survival. Furthermore, the expression of PLK1 was analysed in a human gastric cancer cell line and in deep-frozen gastric cancer tissue using immunoblotting and immunofluorescence techniques. An overexpression of PLK1 was found in 54.1% of analysed carcinomas compared to normal gastric mucosa. This overexpression correlated positively with the tumour stage, the nodal status and patients' age as well as a diffuse growth pattern according to Laurén. PLK1 expression in primary tumour tissue did not considerably differ from PLK1 expression in the corresponding lymph node metastases. 57.3% of carcinomas showed an overexpression of PLK3 with slightly higher PLK3 expression levels in lymph node metastases. PLK3 expression correlated positively with tumour stage and nodal status as well as with PLK1 expression. Univariate survival analysis revealed a significant negative prognostic impact of PLK1 and PLK3 overexpression whereas expression of both isoenzymes had no independent influence on patients' prognosis in multivariate survival analysis. Compared to normal gastric mucosa the analysed gastric cancer cell line and the deep- frozen gastric cancer tissue displayed higher expression levels of PLK1. In conclusion, the present study established an interrelation between the expression of PLK1 and 3 in gastric carcinoma and prognostic factors indicating malignancy as well as patient survival. These results argue for an important role of PLK1 and 3 in oncogenesis of gastric cancer proposing especially the PLK1 molecule as a target of novel chemotherapeutic approaches
