On one hand, the amino acid has been introduced into an oligopeptide chain to allow further transformation with the electron rich double bond, on the other hand, this non-natural amino acid is used as a chiral starting material to construct a range of more complicated amino acids and alkaloids. In this work, a highly flexible and efficient new synthetic route has been developed to generate various optically active C-allylglycine derivatives. Likewise, R and S absolute configurations could be introduced selectively by choosing an appropriate strategy. An auxiliary directed zwitterionic aza-Claisen rearrangement served as the key reaction to obtain unsaturated a-amino acid amides with acceptable yields and high diastereoselectivity. Various N-allyl proline derivatives and a-chloro as well as a-amino acetyl fluorides have been synthesized as suitable starting compounds. The zwitterionic aza-Claisen rearrangement afforded C-allylglycine derivatives with various functional groups reached the nitrogen. The chiral auxiliary could be removed under acidic conditions to generate carboxylic acids and esters in one step, respectively. Furthermore, the chiral auxiliary could be removed to forming C-C bonds by treating the amides with organolithium reagents. Intending to use the rearrangement products for amaryllidaceae alkaloid total syntheses, the introduction of a piperonyl substituent and ring closure via ring-closing metatheses have been investigated.Optisch aktive C-Allylglycin-Derivate sind vielfältig verwendbare Startmaterialien in der organischen Synthese. Sie dienen einerseits als weiter funktionalisierbare Monomere in (Oligo-) Peptiden, andererseits können aus ihnen diverse komplexe Aminosäuren und Alkaloide aufgebaut werden. Optisch aktive C-Allylglycine werden gewöhnlich durch de-novo-Synthesen gewonnen. Das Einführen der chiralen Information erfolgt meist mittels Auxiliar- kontrollierter Reaktionen. In der vorliegenden Arbeit werden N-Allyl-(S)-Prolinderivate als chirale Allylüberträger eingesetzt: Die Reaktion mit Glycinylfluoriden erlaubt den direkten Aufbau von (R)-C-Allylglycinamiden. Das Stickstoffzentrum kann dabei im weiten Rahmen variabel substituiert sein, mindestens ein Akzeptorsubstituent ist jedoch erforderlich. Das Einsatzgebiet der (R)- C-Allylglycine ist auf dem Gebiet der Totalsynthese angesiedelt, hier werden flexible Zugänger zu Crinan und anderen Amaryllidaceae Alkaloiden gesucht. a-N-BOC-Amide können leicht in o-Brom-N-piperonylsysteme transformiert werden. Ein Halogen / Metallaustausch mit darauffolgender Cyclisierung liefert Isochinolone (Abspaltung des Auxiliars in Syntheseschritt). Alternativ kann ein lithiierter Aromat direkt mit dem C-Allylglycinamid umgesetzt werden, die abschließende Auxiliar- Abspaltung liefert ein acyclisches Keton. Die Ketofunktion kann mittels Allylierung in ein 1,2-Diallylsystem überführt werden. Die sich anschließende Ringschluß-Metathese liefert dann ein definiertes Amino-Cyclohexen (anelliert oder isoliert), welches als Schlüsselintermediat für Amaryllidaceae-Alkaloid- Totalsynthesen vom Crinan-Type dienen soll
