Opioide sind bis heute die potentesten Medikamente zur Therapie akuter und chronischer Schmerzen. Ihre Wirksamkeit wird vermittelt durch Interaktion mit Rezeptoren (mu, delta, kappa) im zentralen und peripheren Nervensystem. Die molekulare Klonierung der cDNA der Opioidrezeptoren und pharmakologische Experimente mit Opioiden trugen entscheidend zur Charakterisierung dieser Rezeptoren bei. Die endogenen Liganden der Opioidrezeptoren sind Opioidpeptide, die aus Precursorpeptiden abgespalten werden.
