During this training, we studied interaction of 5-HT6 receptor with β-arrestins and downstream signalling of 5-HT6R, on ERK and mTOR signalling pathway. We provided first evidence of interaction of 5-HT6R with β-arrestins 2 and demonstrated that WAY-208 466 and 5-HT act as agonists on this recruitment, SB-271 act as antagonist and SGS-518 act as inverse agonist. AlphaScreen results from NG 108-15 lysates silenced for β-arrestin 2 revealed decrease in quantity of phosphorylated ERK, leading to conclusion that 5-HT6R could trigger ERK signalling pathway directly through its interactions with β-arrestin 2.
