Bioactive Circular Cystine-rich Peptides: Discovery, Characterization and Application in Target Identification for Drug Discovery

Natürliche Ressourcen wie zum Beispiel Heilpflanzen sind eine vielversprechende Quelle bioaktiver Substanzen für die Arzneistoffentwicklung. Ribosomal synthetisierte und post-translational modifizierte Peptide, zum Beispiel Zyklotide isoliert von Pflanzen, wurden evolutionär optimiert, um zelluläre Rezeptoren oder Enzyme zu modulieren, daher sind sie ein potentieller Startpunkt für die Entwicklung zukünftiger Arzneistoffkandidaten. Eine N- zu C-terminale Ligation mittels einer Peptidbindung und der sogenannten Cystinknoten bestehend aus drei Cystinbrücken stabilisieren Zyklotide. Die daraus resultierende erhöhte Integrität gegenüber chemischen, thermischen und enzymatischen Belastungen ist für die Entwicklung von Zyklotid-basierenden Arzneistoffen von wesentlicher Bedeutung. In der vorgelegten Dissertation präsentiere ich meine wissenschaftlichen Arbeiten im Bereich der Zyklotidforschung. Unter anderem beschäftige mich die Entdeckung neuer Zyklotide und deren analytischen Charakterisierung. Neues methodisches Vorgehen kombinierte Transkriptom- und Proteomanalysen, um einen umfassenden Einblick in die Vielfalt von Zyklotiden der Modelpflanze Viola tricolor und mehreren Pflanzen der Rubiaceae Familie (Rötegewächse) zu gewinnen. Meine weitere wissenschaftliche Tätigkeit befasste sich mit der Entdeckung und der Erforschung neuer Bioaktivitäten. So wurden Pflanzenextrakte von drei Psychotrien und einer Violaceae einer Pharmakologie geführten Isolationsstrategie unterworfen, wodurch ein Peptidinhibitor des Enzymes Prolyloligopeptidase isoliert und chemisch charakterisiert werden konnte. Die naturheilkundliche Verwendung der Pflanze Rinorea dentata ergab Hinweise auf eine ethnomedizinische Anwendung zur uterinen Stimulation. Rinorea Pflanzenextrakte induzierten ex vivo eine myometriale Uteruskontraktion und bioaktive Zyklotide konnten in dieser Pflanze identifiziert werden. Peptide von V. tricolor zeigten eine anti-proliferative Aktivität gegenüber aktivierten T-lymphozyten. Mit Hilfe eines Tiermodels für Multiple Sklerose wurde bewiesen, dass bioaktive Zyklotide eine Eignung als immunsuppressive Arzneistoffe aufweisen. Das Peptid [T20K]kalata B1 war stabil genug, um nach peroraler Gabe sein Wirkungsspektrum entfalten zu können. Eine Methodik zur Ermittlung molekularer Interaktionspartner für zyklische bioaktive Peptide wurde entwickelt und ergab die Familie der 14-3-3 Proteine als Bindungspartner für [T20K]kalata B1. 14-3-3 Proteine übernehmen eine Rolle in der Proliferationsteuerung von Lymphozyten ein, indem sie apoptotische Signaltransduktionswege kontrollieren. Eine direkte Verbindung zwischen der nachgewiesenen molekularen Interaktion und der Wirkung im Tiermodel muss durch nachfolgende Studien belegt werden. Zusammenfassend, befasst sich meine Arbeit mit der Entdeckung und Charakterisierung sowie mit Applikationen von natürlichen bioaktiven Peptiden im Bereich von früher präklinischen Forschung und Arzneistoffentwicklung.Natural products including small molecules, peptides and biopolymers are a versatile source of drug leads. A largely underexplored class of bioactive compounds are ribosomally and post-translationally modified peptides from bacteria, plants or animals. These peptides have been evolutionarily optimized for defense or hunting over millions of years to modulate specific types of enzymes, ion channels and receptors. Hence, nature-derived combinatorial peptide libraries may find application in drug discovery and human medicine. Amongst others, plant derived cyclotides are promising drug candidates. They are cyclic peptides containing a cystine-knot fold, which confers them remarkable stability toward enzymatic, chemical and thermal degradation. In this doctoral thesis I have worked on the discovery and characterization of cyclotides as well as on their application in drug discovery. The coffee and violet plant families were investigated by transcriptomics and peptidomics using analysis at the nucleic acid level from available transcriptome datasets and at the proteome level using mass spectrometry. I have developed a methodology that provides a starting point for next-generation cyclotide discovery and characterization in bioactive extracts from natural sources. Furthermore, I focused on drug discovery and target identification. For example, Rinorea dentata plant extracts, used in South-West Nigerian ethnomedicine were tested in an ethnopharmacology guided discovery approach ex vivo for myometrial contractility and a cyclotide was isolated to provide evidence for the presence of bioactive compounds in the plant species. Cyclotides from several Psychotria and one Viola plant species were used in target-based drug discovery. Pharmacology-guided fractionation enabled the isolation of a cyclotide inhibitor of the human prolyl oligopeptidase. Phenotypic drug discovery screens were performed using Viola tricolor extracts to show an anti-proliferative potency on activated human T-lymphocytes. The suppression of cell proliferation was further explored in vivo in an animal model mediated by auto-reactive T- lymphocytes to highlight a possible therapeutic application for the peptide. The cyclotides bioactivity after oral administration was confirmed reducing the disease progression in a model for multiple sclerosis. Exemplarily, one anti-proliferative cyclotide was used for target deconvolution to elucidate molecular binding sites in the biological context of the reported in vitro and in vivo activity. The family of 14-3-3 proteins was identified as possible targets for [T20K]kalata B1. Generally, 14-3-3 proteins are known to regulate T-cell proliferation by controlling apoptotic pathways in cells. The cyclotides molecular interaction to a disease relevant protein ultimately provides the basis for further studies to investigate their mechanism of action.
