The human genome is hierarchically folded in the three-dimensional nucleus. Pairwise chromatin contacts cluster in discrete chromosomal regions, termed topologically associating domains (TADs). Whether TADs play an essential role in gene expression regulation in evolution and genetic diseases, is analyzed in this thesis by computationally integrating genome-wide contact maps with various data along the linear genome. Thereby, functionally related genes cluster in TADs and share distal regulatory elements to enable coordinated gene expression. TADs are primarily stable during evolution and associate with conserved expression profiles.
