Alterações funcionais induzidas por células trofoblásticas em monócitos infectados por Toxoplasma gondii

Toxoplasma gondii is an intracellular parasite that can infect humans and cause toxoplasmosis. This pathogenesis is potentially severe in immunocompromised individuals and during pregnancy because congenital infection can adversely affect both the gestation and normal development of the embryo. During pregnancy, trophoblast cells constitute an important maternal-fetal barrier and innate immune cells, such as monocytes, concentrate around them. Thus, interactions between trophoblast cells and monocytes are important to maintain the normal course of pregnancy, especially in situations of infection. This study aimed to evaluate if trophoblast cells (BeWo cell line) modulate the functional activity of monocytes (THP-1 cell line) in the presence or absence of T. gondii infection. The perfil of cytokines (MIF, IL-12, IL-6, IL-17A, TGF-β1 e IL-10) secreted by THP-1 and BeWo cells was characterized and both of the cell lines show profile of pro-inflammatory cytokines after the infection. Furthermore, THP-1 cells were stimulated with supernatants of BeWo cells previously infected or not with T. gondii and, after this, THP-1 cells were infected with T. gondii. The secretion of cytokines by THP-1 cells was then evaluated, as well as the susceptibility of these cells to T. gondii infection. The results demonstrated that the supernatant of infected BeWo cells induced non-infected THP-1 cells to secret higher levels of pro-inflammatory cytokines and the supernatant of non-infected BeWo cells decreased the secretion of MIF, increased the secretion of IL-6 and not altered the secretion of IL-12 and IL- 17 by these THP-1 cells. Furthermore, both of the supernatants induced lower secretion of MIF and IL-12 and higher secretion of IL-6 by THP-1 cells infected with T. gondii. The secretion of IL-17 by infected THP-1 cells was higher only with stimuli of infected BeWo cells. The secretion of TGF-β1 by THP-1 cells stimulated with the supernatants of uninfected or infected BeWo cells was down-modulated while the secretion of IL-10, in the same conditions, was up-modulated. In addition, THP-1 cells stimulated, principally with the supernatants of previously infected BeWo cells, were more resistant to T. gondii proliferation. THP-1 cells treated with rIL-6 were less permissive to T. gondii proliferation and in the absence of this cytokine, this parasite proliferation increased. Thus, the secretion of IL-6 by THP-1 cells is involved in the control of T. gondii infection. Additionally, the secretion of IL- 6 and MIF by BeWo cells, but not the secretion of TGF-β1, was important to control the parasitism in THP-1 cells. Esta doença é potencialmente grave em indivíduos imunocomprometidos e durante a gestação, uma vez que a infecção congênita pode comprometer a gestação e o desenvolvimento normal do embrião. Durante a gestação, células trofoblásticas constituem uma importante barreira materno-fetal e células imunes inatas, como os monócitos, se concentram ao redor dessas células trofoblásticas. Assim, interações entre células trofoblásticas e monócitos são importantes para manter o curso gestacional, especialmente em situações de infecção. Este estudo teve como objetivo avaliar se células trofoblásticas (linhagem BeWo) modulam a atividade funcional de monócitos (linhagem THP-1) na presença ou ausência de infecção por T. gondii. Inicialmente caracterizou-se o perfil de citocinas (MIF, IL-12, IL-6, IL-17A, TGF-β1 e IL-10) secretadas por células THP-1 e BeWo não-infectadas e infectadas por T. gondii. Verificou-se que ambas as linhagens celulares exibem perfil de citocinas pró-inflamatórias quando infectadas. Posteriormente, células THP- 1 foram estimuladas com sobrenadantes de células BeWo não-infectadas ou infectadas com T. gondii e, após esse estímulo, células THP-1 foram infectadas com T. gondii. A secreção de citocinas por células THP-1 foi avaliada, bem como a susceptibilidade dessas células à infecção por T. gondii. Os resultados demonstraram que o sobrenadante de células BeWo infectadas induziu células THP-1 não-infectadas a secretarem níveis maiores de citocinas próinflamatórias e o sobrenadante de células BeWo não-infectadas diminuiu a secreção de MIF, aumentou a secreção de IL-6 e não alterou a secreção de IL-12 e IL-17 por estas células THP- 1. Além disso, os dois diferentes sobrenadantes foram capazes de induzir secreção menor de MIF e IL-12 e secreção maior de IL-6 por células THP-1 infectadas com T. gondii. A secreção de IL-17 por células THP-1 infectadas foi maior apenas com estímulo de células BeWo infectadas. A secreção de TGF-β1 por células THP-1 estimuladas com sobrenadante de células BeWo não-infectadas ou infectadas diminuiu, enquanto que a secreção de IL-10, nas mesmas condições, aumentou. Além disso, células THP-1 estimuladas, principalmente, com sobrenadante de células BeWo infectadas, foram mais resistentes à proliferação de T. gondii. Células THP-1 tratadas com rIL-6 foram menos permissivas à proliferação de T. gondii e na ausência desta citocina, a proliferação parasitária foi maior. Portanto, a secreção de IL-6 por células THP-1 está envolvida no controle da infecção por T. gondii. Adicionalmente, a secreção de IL-6 e MIF por células BeWo, mas não a secreção de TGF-β1, foi importante para controlar o parasitismo em células THP-1. Em conjunto, estes resultados sugerem que células trofoblásticas são hábeis em modular a atividade de monócitos e este processo pode estar associado à manutenção da gestação
