Phenotyping and genotyping of onion populations with different levels of inbreeding

The production of onion hybrids depends on the development of genetically pure lineages. Traditionally, the pedigree method (applying successive self-fertilization in individuals) guarantees obtaining quickly the endogenous lines. However, the onion undergoes a strong process of inbreeding depression when self-fertilized for several generations, such as many other species of cross-pollination. Depression by inbreeding reduces plant vigor, bulb size and seed production, increasing the cost to produce the hybrid seed. Researchers have shown that cross-linking between a few plants promotes the reduction of depression by inbreeding in natural populations. Based on the same principle and taking in consideration the populations submitted to the selection of superior individuals, the objective of this paper is to elucidate if there is any effect of depression by inbreeding segregating lines of onion obtained by self-fertilization and by cross-linking of two and three plants. Therefore, we used 33 populations of onion and designed treatments in different ways of obtaining the populations (self-fertilization of one plant [A1], intercrossing of two [B2] and three plants [C3]), to study 11 different genetic backgrounds, grouped according to low or medium inbreeding. We evaluate throughout two consecutive years (2014 and 2015) the agronomic characteristics in the vegetative and reproductive phases of the crop, and conducted analysis with RAPDs and SSRs. The experiments were conducted at the experimental station of Bayer Vegetable Seeds in Uberlândia/MG. Plant vigor suffered the depressed by inbreeding in the early generations with greater heterozygosity. Similarly, the contrast between the self-fertilized compared to the two-or three-plant crossbred was more pronounced in the populations with lower endogamic level. The intercrossing of three plants reflected in larger production of seeds per plot in populations with low endogamic level. Changing the method of conducting the generations through the sib of plants provided greater divergence between the genotypes. Important characteristics such as bulb production per plot, seed survival, resistance to purple blotch (fungus Alternaria porri), and seed production by umbel, that can be used in the estimation of divergence in the selection of segregating onion lines. We observed a positive and significant genotypic correlation of 0.92 for the percentage of seed survival, and 0.82 for the resistance to purple blotch, with seed production per umbel. These results indicates that the weight of seeds per umbel can be increased through selection for resistance to purple blotch and percentage of seed survival. Furthermore, we concluded the depression by inbreeding could not be interpreted as due to homozygous effects for deleterious mutations, and that microsatellites markers may be useful for detecting variability within the Brazilian onion germplasm. These results suggest that sibs of two and three plants can be used to reduce the inbreeding effect in onion, and should start in low endogamous populations, for the best use of the method.CAPES - Coordenação de Aperfeiçoamento de Pessoal de Nível SuperiorTese (Doutorado)A produção de híbridos de cebola depende do desenvolvimento de linhagens geneticamente puras. Tradicionalmente, o método de ‘pedigree’ (onde se aplica sucessivas autofecundações em nível de indivíduos), é o que garante uma rápida obtenção de linhas endogâmicas. No entanto como muitas espécies de polinização cruzada, a cebola sofre um forte processo de depressão por endogamia quando autofecundadas por várias gerações. A depressão por endogamia reduz o vigor da planta, o tamanho do bulbo e sua produção de sementes, o que acaba onerando o custo de produção da semente híbrida. Sabe-se que o intercruzamento entre poucas plantas promove a diminuição da depressão por endogamia em populações naturais. Baseado no mesmo princípio, ainda que, se tratando de populações submetidos à seleção de indivíduos superiores, este trabalho tem o objetivo de elucidar se existe algum efeito de depressão por endogamia em linhas segregantes de cebola, obtidas pela autofecundação e pelo intercruzamento de duas e três plantas. Para isso, utilizou-se para o estudo 33 populações de cebola, os tratamentos constituíram-se das diferentes formas de obtenção das populações (autofecundação de uma planta [A1], intercruzamento de duas [B2] e três plantas [C3]) em 11 diferentes backgrounds genéticos agrupados de acordo com o nível baixo ou médio de endogamia. Foram avaliadas em dois anos consecutivos (2014 e 2015), características agronômicas na fase vegetativa e reprodutiva da cultura, além de análises com marcadores RAPD’s e SSR’s. Os experimentos foram realizados na estação experimental da Bayer Vegetable Seeds em Uberlândia/MG. O vigor de plantas sofreu efeito de depressão por endogamia em gerações iniciais com maior heterosigosidade; assim como o contraste entre indivíduos autofecundados comparativamente aos advindos do intercruzamento de duas ou três plantas foi mais pronunciado nas populações com menor nível endogâmico. O intercruzamento de três plantas refletiu em maior produção de sementes/parcela nas populações com baixo nível endogâmico. A alteração no método de condução das gerações, através do sib de plantas, proporcionou maior divergência entre os genótipos. Características importantes, como produção de bulbos por parcela, pegamento de sementes, resistência à Alternária e produção de sementes por umbela, que podem ser usadas nas estimativas da divergência na seleção de linhas segregantes de cebola. Observou-se ainda, correlação genotípica positiva e significativa de 0,92 e 0,82, da porcentagem de pegamento de sementes e resistência a alternária, respectivamente com produção de sementes por umbela, indicando que o peso de sementes por umbela pode ser aumentado através da seleção para resistência a alternária e porcentagem de pegamento de sementes. Por fim, conclui-se que a depressão por endogamia não pode ser prontamente interpretada como devida a efeitos de homozigoticidade para mutações deletérias e que os microssatélites usados podem ser úteis para detectar variabilidade no germoplasma brasileiro de cebola. Os resultados sugerem que os sibs de duas e três plantas podem ser utilizados visando reduzir o efeito endogâmico em cebola, devendo iniciar em populações com nível baixo de endogamia para maior proveito do método
