Psychostimulant sensitization is a well-described phenomenon wherein behavioral responses to stimulant drugs like amphetamine and cocaine become stronger with subsequent doses. These behavioral changes co-vary with neurochemical changes within mesolimbic pathways, including increases to dopamine transmission and diverse alterations to glutamate signaling. It has been proposed that these sensitized responses could contribute to a number of psychiatric conditions, with particularly compelling links to substance use disorders and schizophrenia.
