Background: Breast cancer is the most common type of cancer in women, and a major cause of premature death still today. The PI3K-Akt-mTOR signaling (in short: PI3K signaling) pathway is a growth and pro-survival pathway, and is frequently deregulated in breast cancer due to amplifications, mutations and deletions of the genes encoding these important signaling hubs. PTEN inhibits the activation of Akt, and acts as an endogenous tumor suppressor by negatively regulating PI3K signaling. Loss of PTEN protein expression is frequently observed in breast cancer, which could contribute to hyperactivation of the PI3K pathway.
