The mitochondrial Permeability Transition Pore (PTP) has been characterized in mammals as a Ca2+-activated channel whose opening allows solutes of molecular mass ≤ 1.5 kDa to equilibrate across the inner membrane, potentially causing matrix swelling, rupture of the outer membrane and release of pro-apoptotic factors. Together with Ca2+, the onset of PTP opening is favored by oxidative stress, membrane depolarization and Pi; conversely, acidic pH, adenine nucleotides and Mg2+ are the major physiological desensitizers.
