The site of uncoupling of neuronal nitric oxide synthase

Die NOS produziert unter bestimmten Bedingungen O2- anstelle von NO. Dieses Phänomen wird als Entkopplung bezeichnet und tritt auf, wenn das Substrat Arginin und/oder der Cofaktor BH4 fehlt. Mögliche Quellen von O2 sind die Flavine in der Reduktase-Domäne und das Häm in der Oxygenase-Domäne. BH4 kann eine entkoppelte Reaktion entweder durch Dimerstabilisierung oder durch Reduktion des Oxyferro-Komplexes verhindern. Im ersten Fall ist das Enzym durch Monomerisierung nicht in der Lage, Elektronen auf die andere Untereinheit zu übertragen, stattdessen reduzieren Flavine O2 zu O2-. Im letzteren Fall verhindert BH4 die Entkopplung durch schnelle Reduktion des eher instabilen Fe(II)O2-Komplexes. Im Rahmen der vorliegenden Diplomarbeit wird untersucht, ob die Entkopplung in der Reduktase oder Oxygenase-Domäne stattfindet. Entkopplung erfolgte in Abwesenheit von Arg und/oder BH4, jedoch nicht in Abwesenheit von Ca2+ oder CaM (ermöglicht Elektronentransport von der Reduktase-Domäne zum Häm). Darüber hinaus spiegelt die Entkopplung in Gegenwart von NOS-Inhibitoren das Ausmaß wider, in dem diese Verbindungen eine Häm-Reduktion erlauben. Das durch FPLC und LT-PAGE bestimmte Monomer/Dimer-Gleichgewicht zeigte keine Korrelation mit der Entkopplung.Zusammenfassend legt die vorliegende Studie nahe, dass Entkopplung am Häm in der Oxygenasedomäne auftritt und dass keine direkte Verbindung zwischen Monomerisierung und Entkopplung besteht. Der Cofaktor BH4 verhindert die Entkopplung als ein 1-Elektron-Donor.Under some conditions, NOS produces O2- instead of NO. This phenomenon is referred to as uncoupling and occurs when the substrate arginine and/or the cofactor BH4 is omitted. Potential sources of O2- are the flavins in the reductase domain and the heme in the oxygenase domain. BH4 prevents uncoupling either by dimer stabilization or by reduction of the oxyferrous complex. In the first case the enzyme is through monomerization unable to transfer electrons to the other subunit, while still allowing reduction of O2 to O2- by the flavins. In the latter case BH4 prevents uncoupling by fast reduction of the rather unstable Fe(II)O2 complex.The current thesis was aimed to investigate where in the enzyme uncoupling takes place.Uncoupling occurred in the absence of Arg and/or BH4, but not in the absence of Ca2+ or CaM (which is required for electron transfer from the reductase domain to the heme). Furthermore, uncoupling in the presence of NOS inhibitors reflected the extent to which these compounds allowed heme reduction. The monomer/dimer equilibrium determined by FPLC and LT-PAGE showed no correlation with uncoupling.In summary, the current study demonstrate that uncoupling occurs at the heme in the oxygenase domain and that the NOS monomer is not involved.
