Signaling pathways and metabolism of the endocannabinoid anandamide in human brain

Seit der Klonierung des Cannabinoid-1-Rezeptors (CB1) im Jahre 1990 und der Isolierung des ersten Liganden Anandamid zwei Jahre später gelten umfangreiche Bemühungen der Charakterisierung des endogenen Cannabinoidsystems. Vorliegende Arbeit untersucht im menschlichen Gehirn mittels in situ Hybridisierung die mRNA-Expression der wichtigsten Enzyme des Anandamid-Metabolismus NAPE-PLD und FAAH. Des Weiteren wird durch Immunhistochemie, in situ Hybridisierung und RT-PCR das Verteilungsmuster des CB1-Rezeptors beschrieben. Es konnte gezeigt werden, dass sowohl CB1 als auch NAPE-PLD und FAAH in nahezu jeder der acht untersuchten Hirnregionen in unterschiedlicher Stärke exprimiert werden. Die quantitative Auswertung der Expressionsrate einzelner Zellen erbrachte zahlreiche neue Erkenntnisse über den Stoffwechsel von Anandamid in lokalen und überregionalen neuronalen Netzwerken.Since the cloning of the cannabinoid-1-receptor (CB1) in 1990 and the isolation of its first natural ligand anandamide two years later great scientific effort has been devoted to the characterization of the endocannabinoid system. Our study examines the mRNA expression of the two pivotal enzymes in cannabinoid metabolism NAPE-PLD and FAAH in human brain using in situ hybridization. Furthermore we determined the expression patterns of the CB1 receptor by immunohistochemistry, in situ hybridization and RT-PCR. It could be shown that both CB1 and the enzymes NAPE-PLD and FAAH are expressed in varying intensity in almost any of the eight major brain regions studied. Quantification of labeling density in single cells revealed many new insights into anandamide metabolism in local and supraregional neuronal networks
